- Alternative sources of n-3 long-chain polyunsaturated fatty acids in marine microalgaePublication . Martins, Dulce Alves; Custodio, Luisa; Barreira, Luísa; Pereira, Hugo; Ben-Hamadou, Radhouan; Varela, João; Abu-Salah, Khalid M.The main source of n-3 long-chain polyunsaturated fatty acids (LC-PUFA) in human nutrition is currently seafood, especially oily fish. Nonetheless, due to cultural or individual preferences, convenience, geographic location, or awareness of risks associated to fatty fish consumption, the intake of fatty fish is far from supplying the recommended dietary levels. The end result observed in most western countries is not only a low supply of n-3 LC-PUFA, but also an unbalance towards the intake of n-6 fatty acids, resulting mostly from the consumption of vegetable oils. Awareness of the benefits of LC-PUFA in human health has led to the use of fish oils as food supplements. However, there is a need to explore alternatives sources of LC-PUFA, especially those of microbial origin. Microalgae species with potential to accumulate lipids in high amounts and to present elevated levels of n-3 LC-PUFA are known in marine phytoplankton. This review focuses on sources of n-3 LC-PUFA, namely eicosapentaenoic and docosahexaenoic acids, in marine microalgae, as alternatives to fish oils. Based on current literature, examples of marketed products and potentially new species for commercial exploitation are presented.
- Application and demonstration of the ecohydrology approach for the sustainable functioning of the Guadiana estuary (South Portugal)Publication . Chícharo, Luís; Ben-Hamadou, Radhouan; Araújo Amaral, Ana Margarida; Range, Pedro; Mateus, Carmen; Piló, David; Marques, Rute; Morais, P.; Teodosio, MariaThe implementation of the UNESCO demosite at the Guadiana estuary represents an excellent opportunity to demonstrate how to adapt, mitigate and restore functioning of estuaries and coastal areas impacted by dams and climatic changes, using the Ecohydrology approach. Worldwide estuaries are suffering similar anthropogenic impacts that affect ecosystem functioning, biodiversity and resources. Therefore, the mesocosms experiments at the Guadiana Ecohydrology demosite constitute a collection of case studies, whose results can be used for testing solutions for the sustainable management of other estuarine systems. We applied the Ecohydrology concepts and methods to test: a) the usefulness of freshwater inflow pulses to regulate biodiversity and to control microalgal blooms , by regulating nutrient ratios and thus enhancing the bottom-up control of water quality; b) the usefulness of using the feeding ability of bivalve assemblages to control microalgal blooms (top-down control of water quality, and; c) the role of salt marsh plants (Salicornia ramosissima) to reduce excessive concentrations of nutrients and control estuarine water quality. Results show that the interplay between organisms (bivalves, plants) and hydrological factors is a key for improving water quality and sustaining biodiversity and the good ecological status of this estuarine ecosystem.
- Are tidal lagoons ecologically relevant to larval recruitment of small pelagic fish? An approach using nutritional condition and growth ratePublication . Chicharo, Maria Alexandra; Araújo Amaral, Ana Margarida; Faria, Ana Margarida da Silva; Morais, R.; Mendes, C.; Piló, David; Ben-Hamadou, Radhouan; Chícharo, LuísThere are numerous studies dealing with larvae of Small Pelagic Fish (SPF), but only a few have actually addressed advanced larval phases. Temperate coastal lagoons are particularly understudied, due to the absence of standard method to capture advanced larval fish in these near shore shallow habitats. Accordingly, this study aims to describe abundances, nutritional condition and in situ growth of postflexion (SPF) from the Ria Formosa, a tidal coastal lagoon in southern Portugal. The nutritional condition and in situ growth were determined through cohort analysis and standardized RNA:DNA ratio (sRD), complemented with feeding incidence (gut content) and fatty acids (FAs, trophic biomarkers) of postflexion larvae sampled sequentially with light traps, from spring 2005 to summer 2006. Simultaneously, environmental parameters such as water temperature, salinity, dissolved oxygen and chlorophyll a were measured. Post-larvae of SPF were captured through the year in important numbers. The dominant species were Sardina pilchardus (50.7%), Engraulis encrasicolus and Atherina presbyter (11.4%). These results are distinct from those based on adult/juvenile surveys or early planktonic phases in the Ria Formosa where clupeiformes were occasionally reported. Sardines were captured mainly in winter, spring and early summer and anchovies mainly during summer and autumn. Sand smelt, a resident species, was present throughout the year. In the early summer, the three species were present and during this period some diet overlapping occurred, the feeding incidence of the clupeiformes was very low, but atherinids always exhibited full guts reflecting the different gut morphology or indicating different lifecycle strategies. The bulk of the diet was mollusks, crustaceans and appendicularians, for sand smelts, sardines and anchovies, respectively. The results of FA analyses showed some contribution of phytoplankton to SPF in the area. All SPF exhibited higher condition (sRD) and growth rates in summer, which are explained by the adequate temperature and higher planktonic productivity. This is especially relevant because the successful development of postflexion larvae in these nursery areas largely determines the successful recruitment to adult fish populations.
- Biological activities and chemical composition of methanolic extracts of selected Autochthonous microalgae strains from the Red SeaPublication . Pereira, Hugo; Custodio, Luisa; Rodrigues, Maria Joao; Bruno De Sousa, Carolina; Oliveira, Marta; Barreira, Luísa; Neng, Nuno da Rosa; Florencio Nogueira, Jose Manuel; Alrokayan, Salman A.; Mouffouk, Fouzi; Abu-Salah, Khalid M.; Ben-Hamadou, Radhouan; Varela, JoãoFour lipid-rich microalgal species from the Red Sea belonging to three different genera (Nannochloris, Picochlorum and Desmochloris), previously isolated as novel biodiesel feedstocks, were bioprospected for high-value, bioactive molecules. Methanol extracts were thus prepared from freeze-dried biomass and screened for different biological activities. Nannochloris sp. SBL1 and Desmochloris sp. SBL3 had the highest radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl, and the best copper and iron chelating activities. All species had potent butyrylcholinesterase inhibitory activity (>50%) and mildly inhibited tyrosinase. Picochlorum sp. SBL2 and Nannochloris sp. SBL4 extracts significantly reduced the viability of tumoral (HepG2 and HeLa) cells with lower toxicity against the non-tumoral murine stromal (S17) cells. Nannochloris sp. SBL1 significantly reduced the viability of Leishmania infantum down to 62% (250 mu g/mL). Picochlorum sp. SBL2 had the highest total phenolic content, the major phenolic compounds identified being salicylic, coumaric and gallic acids. Neoxanthin, violaxanthin, zeaxanthin, lutein and -carotene were identified in the extracts of all strains, while canthaxanthin was only identified in Picochlorum sp. SBL2. Taken together, these results strongly suggest that the microalgae included in this work could be used as sources of added-value products that could be used to upgrade the final biomass value.
- Calcification, growth and mortality of juvenile clams Ruditapes decussatus under increased pCO(2) and reduced pH: variable responses to ocean acidification at local scales?Publication . P.; Chícharo, LuísWe investigated the effects of ocean acidification on juvenile clams Ruditapes decussatus (average shell length 10.24 mm) in a controlled CO2 perturbation experiment. The carbonate chemistry of seawater was manipulated by diffusing pure CO2, to attain two reduced pH levels (by −0.4 and −0.7 pH units), which were compared to unmanipulated seawater. After 75 days we found no differences among pH treatments in terms of net calcification, size or weight of the clams. The naturally elevated total alkalinity of local seawater probably contributed to buffer the effects of increased pCO2 and reduced pH. Marine organisms may, therefore, show diverse responses to ocean acidification at local scales, particularly in coastal, estuarine and transitional waters, where the physical–chemical characteristics of seawater are most variable. Mortality was significantly reduced in the acidified treatments. This trend was probably related to the occurrence of spontaneous spawning events in the control and intermediate acidification treatments. Spawning, which was unexpected due to the small size of the clams, was not observed for the pH −0.7 treatment, suggesting that the increased survival under acidified conditions may have been associated with a delay in the reproductive cycle of the clams. Future research about the impacts of ocean acidification on marine biodiversity should be extended to other types of biological and ecological processes, apart from biological calcification.
- Comparative study of shell shape and muscle scar pigmentation in the closely related cupped oysters Crassostrea angulata, C-gigas and their reciprocal hybridsPublication . Based on larval shell morphology, experimental hybridization, allozymes and nuclear DNA studies several authors have considered these two taxa as being synonymous. However, mitochondrial data showed clear genetic differences between the two taxa. In addition, microsatellite- based studies and cytogenetic studies have also provided evidence that supports their differentiation. Considerable differences have also been observed at the phenotypic level in terms of growth rate and ecophysiological parameters. In the present study, C. angulata from Sado estuary ( Portugal) and C. gigas from Seudre estuary ( France) were collected and factorial crosses were performed. Juveniles of the different progenies were reared in Ria Formosa ( Portugal) under common conditions to determine if they exhibited differences in shell shape and in pigmentation of the adductor muscle scar. Significant morphometric differences between C. angulata and C. gigas progenies were indicated by univariate and multivariate analyses. Univariate analysis of size- adjusted shell measurements revealed significant differences between the two taxa for shell depth, muscle scar height, and length of ligamental area. Both reciprocal hybrids showed intermediate morphometric characters between parental lines. In addition, significant differences were also observed between C. angulata and C. gigas progenies in terms of pigmentation of adductor muscle scar. C. angulata and both reciprocal hybrid progenies showed highly pigmented adductor muscle scars whereas in C. gigas progeny the pigmentation was lighter. The differences in shell shape and muscle scar pigmentation observed in the present study support the distinction of the two taxa.
- Cytogenetics of Bolinus brandaris and phylogenetic inferences within the Muricidae (Mollusca: Gastropoda)Publication . Leitao, Alexandra; Vasconcelos, Paulo; Ben-Hamadou, Radhouan; Gaspar, Miguel; Barroso, Carlos M.; Ruano, FranciscoThe purple dye murex, Bolinus brandaris (Linnaeus, 1758), is a muricid gastropod common throughout the Mediterranean and along the Moroccan and Portuguese Atlantic coasts. In the present study, we confirmed the diploid chromosome number of 2n = 70 for this species, and established for the first time the karyotype, which comprised 12 metacentric, 15 submetacentric and eight subtelocentric chromosome pairs. To facilitate cytotaxonomic comparisons, we carried out a comparative karyological analysis through multidimensional scaling between B. brandaris and three other 2n = 70 muricid species (Hexaplex trunculus, Ocenebra erinaceus, and Stramonita haemastoma) for which chromosomal measurements have been previously published. The interpretation of the ideograms and the statistical analysis highlighted the closest similarity of B. brandaris and H. trunculus compared to S. haemastoma and O. erinaceus. Indeed, B. brandaris and H. trunculus showed the smallest dissimilarities both for relative length and arm ratio, with O. erinaceus presenting intermediate values, whereas the highest dissimilarities were found between H. trunculus and S. haemastoma for both data. The karyotypes of B. brandaris and H. trunculus (subfamily Muricinae) presented the highest proportions of metacentric chromosomes compared to the other two muricids analysed, suggesting that those karyotypes could be considered primitive within the 2n = 70 Muricidae studied so far. - Diet and feeding intensity of sardine Sardina pilchardus: correlation with satellite-derived chlorophyll dataPublication . Garrido, Susana; Ben-Hamadou, Radhouan; Oliveira, P. B.; Cunha, M. D.Spatio-temporal variability of the diet of sardine Sardina pilchardus off Portugal was examined through analysis of the stomach contents of fish collected every 14 d from the west and south of Portugal during 2003/2004. Dietary composition of the modal sardine length class was assessed by determining the frequency of occurrence and carbon content of identified prey, and these 2 parameters were combined to estimate a modified index of relative importance of prey (mIRI). The most important prey for sardines were zooplankton, comprising crustacean eggs, copepods, decapods, cirripedes and fish eggs, dinoflagellates and diatoms (particularly the toxin-producer genus Pseudo-nitzschia), which together accounted for >90% of the estimated dietary carbon. Dietary seasonality was similar for both areas, except that the contribution of phytoplankton was higher for fish from the west Portuguese coast, where upwelling events are stronger and recurrent during spring and summer months. The predominance of prey <750 μm in sardine diet suggests that filter feeding is the dominant feeding mode used in the wild. Feeding intensity was similar for both sexes and for fish of different length classes and was higher on the west coast than in the south, which is probably related to the higher productivity of the west coast. Although there was high inter-annual variability in feeding intensity, this parameter was highest for both areas during spring and winter months. Temporal variability in satellite-derived chlorophyll a matched the temporal variability in the dietary contribution by phytoplankton and of sardine feeding intensity, suggesting further investigation of the potential use of satellite-derived chlorophyll a data as a proxy for sardine feeding intensity.
- Effect of maternal fat reserves on the fatty acid composition of sardine (Sardina pilchardus) oocytesPublication . Garrido, Susana; Rosa, R.; Ben-Hamadou, Radhouan; Cunha, M. D.We compared the fatty acid (FA) composition of the muscle and gonads of female Iberian sardines with hydrated oocytes collected during the 2002/03 spawning season off southern Portugal (November and February) and off western Portugal (February). Sardine condition and total FA concentration in the muscle decreased between the two sampling dates, while the gonadosomatic index was similar between samples. Total monounsaturated FA concentrations in sardine gonads were different for the three samples while saturated and polyunsaturated FA concentrations were similar. Significant linear relations were found between FA concentrations in female muscle and oocytes, including eicosapentaenoic acid (EPA; 20:5n−3) and arachidonic acid (AA; 20:4n−6), both being essential for normal larval development. The concentration of docosahexaenoic acid (DHA; 22:6n−3) in oocytes was independent on muscle concentration, probably resulting from its selective transfer to the oocytes. The EPA/ DHA ratio was highly conserved in sardine tissues, while DHA/AA and EPA/AA ratios varied significantly between samples. These results indicate that the FA content of eggs produced by sardines varies throughout the spawning season, egg FA concentrations decreasing as females lose condition, and FA composition also shows spatial variability. Both types of variability may have a significant impact on egg quality, particularly on the amount of reserves available to larvae affecting their resistance to starvation, and the appropriate FA composition required for normal growth.
- Effect of temperature on the growth, survival, development and foraging behaviour of Sardina pilchardus larvaePublication . A.; Re, P.; Santos, A.M.P.The effect of water temperature on the growth, survival, development and foraging behaviour of European sardine Sardina pilchardus larvae was examined in the laboratory. First, the capability of early sardine larvae to cope with starvation was assessed at temperatures from 10 to 22 degrees C. Second, we examined under ad libitum feeding conditions and across the range of temperatures experienced by sardines during spawning along the Atlanto-Iberian coast (13-17 degrees C) the ontogenetic changes in growth, survival and foraging behaviour of sardine larvae. Unfed larvae had similar maximum survival times (11-12 d post hatching, dph) from 13 to 15 degrees C, but the survival time was significantly shorter at the coldest and warmest temperatures tested. The survival of exogenously feeding larvae increased with temperature, but younger endogenously feeding larvae had higher survival at colder temperatures. The cumulative mortality after 25 dph, however, was similar at the 3 temperatures. Not only larval growth rate increased with increasing temperature, but ontogenetic development also occurred sooner and at smaller sizes. Notochord flexion, which is a developmental milestone for fish, occurred 10 d earlier at 17 rather than at 13 degrees C. The time spent swimming and the foraging behaviour (orientations to prey, feeding strikes and successful capture) significantly increased throughout the ontogeny and with temperature. This study highlights how even modest changes in spawning temperature can lead to large changes in the survival and growth of larval sardine. This study also reveals some of the mechanisms whereby inter-annual and seasonal variability in temperature can have significant ecological impacts at the population level.
