T.; Jorge Parola, A.; Basilie, NunoThe discovery of stimuli-responsive high affinity host-guest pairs with potential applications under biologically relevant conditions is a challenging goal. This work reports a high-affinity 1:1 complex formed between cucurbit[8]uril and a water-soluble photochromic diarylethene derivative. It was found that, by confining the open isomer within the cavity of the receptor, a redshift in the absorption spectrum and an enhancement of the photocyclization quantum yield from phi=0.04 to phi=0.32 were induced. This improvement in the photochemical performance enables quantitative photocyclization with visible light that, together with the near-infrared light-induced ring-opening reaction and the 100-fold selectivity for the closed isomer, confirms this as an outstanding light-responsive affinity pair.
- Effect of in vitro gastrointestinal digestion on the total phenolic contents and antioxidant activity of wild Mediterranean edible plant extractsPublication . Gonçalves, Sandra; Moreira, E.; Andrade, Paula B.; Valentao, Patricia; Romano, AnabelaThe recent interest in wild edible plants is associated with their health benefits, which are mainly due to their richness in antioxidant compounds, particularly phenolics. Nevertheless, some of these compounds are metabolized after ingestion, being transformed into metabolites frequently with lower antioxidant activity. The aim of the present study was to evaluate the influence of the digestive process on the total phenolic contents and antioxidant activity of extracts from four wild edible plants used in the Mediterranean diet (Beta maritima L., Plantago major L., Oxalis pes-caprae L. and Scolymus hispanicus L.). HPLC-DAD analysis revealed that S. hispanicus is characterized by the presence of caffeoylquinic acids, dicaffeoylquinic acids and flavonol derivatives, P. major by high amounts of verbascoside, B. maritima possesses 2,4-dihydroxybenzoic acid, 5-O-caffeoylquinic acid, quercetin derivatives and kaempferol-3-O-rutinoside, and O. pes-caprae extract contains hydroxycinnamic acids and flavone derivatives. Total phenolic contents were determined by Folin-Ciocalteu assay, and antioxidant activity by the ABTS, DPPH, ORAC and FRAP assays. Phenolic contents of P. major and S. hispanicus extracts were not affected by digestion, but they significantly decreased in B. maritima after both phases of digestion process and in O. pes-caprae after the gastric phase. The antioxidant activity results varied with the extract and the method used to evaluate the activity. Results showed that P. major extract has the highest total phenolic contents and antioxidant activity, with considerable values even after digestion, reinforcing the health benefits of this species.
- Combination of hyaluronic acid and PLGA particles as hybrid systems for viscosupplementation in osteoarthritisPublication . HA is generally supplied as an injection commonly reported as painful, with strong limitations after treatment. In this study, an alternative delivery system was constructed based on HA hydrogel and poly(lactic-co-glycolic acid) (PLGA) particles with oleic acid. Development studies included the determination of particle toxicity, hemolytic activity, in vitro and in vivo anti-inflammatory activity using macrophages and a murine model, respectively. This study showed that empty PLGA particles presented a mean size of 373 nm, while particles containing HA and oleic acid showed a marked particle size increase. The HA association efficiency was of 73.6% and 86.2% for PLGA particles without and with oleic acid, respectively. The in vitro HA release from PLGA particles revealed a sustained profile. Particles showed a good in vitro cell compatibility and the risk of hemolysis was less < 1%, ensuring their safety. The in vivo anti-inflammatory study showed a higher inhibition for HA-loaded PLGA particles when compared to HA solution (78% versus 60%) and they were not different from the positive control, clearly suggesting that this formulation may be a promising alternative to the current HA commercial dosage form.
- The Atlantic blue crab Callinectes sapidus Rathbun, 1896 expands its non-native distribution into the Ria Formosa lagoon and the Guadiana estuary (SW-Iberian Peninsula, Europe)Publication . Morais, Pedro; Gaspar, Miguel; Garel, Erwan; Baptista, Vânia; Cruz, Joana; Cerveira, Inês; Leitão, Francisco; Teodosio, MariaThe Atlantic blue crab Callinectes sapidus Rathbun, 1896 is native in the western Atlantic, however it is a non-indigenous species across Europe since 1900, among other world regions. In this paper, we report the first occurrences of this species in the Ria Formosa lagoon and in the Guadiana estuary (SW-Iberian Peninsula, Europe) which occurred in 2016 and July 2017, respectively. We hypothesize that the introduction of this species into these ecosystems might be due to the expansion of the Guadalquivir estuary population through natural processes (larval advection, active movement), or due to unintended introduction events after being transported aboard fishing boats, or, less likely, through ballast water. Changes in Guadiana's river flow after the construction of the Alqueva dam might also explain the presence of another non-indigenous species in the Guadiana estuary. The hypotheses presented, regarding the introduction of the Atlantic blue crab into these ecosystems and of its co-occurrence with other decapod species, are framed in a broader context to serve as a future research framework. The use of the Atlantic blue crab as a new fishing resource is also proposed, namely if it is to be used exclusively by local communities and if no deleterious impacts upon other fisheries and the ecosystem occur from this new fishery.
- Pitaia: perspetivas e dificuldades de uma “nova” culturaPublication . Trindade, Ana Rita; Reis, Adélia; Sabbo, Luís; Trindade, Diamantino; Paiva, Paulo; Duarte, AmilcarNo Algarve, onde existem condições edafoclimáticas favoráveis ao cultivo de algumas espécies frutícolas exóticas, oriundas de climas tropicais ou subtropicais, surge o interesse no cultivo da pitaia. Esta cultura apresenta altas produções em alguns países e é adequada para terrenos de pequenas dimensões. Ela acaba por satisfazer também a necessidade de diversificar a fruticultura da região. Devido à sua aparência exótica e ao seu alto valor nutritivo, a pitaia tem sido cada vez mais procurada em diversos países, principalmente pelos mercados asiáticos e europeus. Para desenvolver a cultura da pitaia no Algarve foi constituído um grupo operacional cujo objetivo principal visa inovar ao nível das tecnologias de produção para a pitaia vermelha, testando a sua produtividade, rusticidade e qualidade dos frutos. Começou-se por fazer um levantamento da situação desta cultura em Portugal. Estão também a ser instalados vários campos de ensaio/recolha de dados, sobre várias espécies de pitaia vermelha.
- Variations of the Shifting Lemma and Goursat categoriesPublication . Gran, Marino; Rodelo, Diana; Nguefeu, Idriss TchoffoWe prove that Mal'tsev and Goursat categories may be characterized through variations of the Shifting Lemma, that is classically expressed in terms of three congruences R, S and T, and characterizes congruence modular varieties. We first show that a regular category C is a Mal'tsev category if and only if the Shifting Lemma holds for reflexive relations on the same object in C. Moreover, we prove that a regular category C is a Goursat category if and only if the Shifting Lemma holds for a reflexive relation S and reflexive and positive relations R and T in C. In particular this provides a new characterization of 2-permutable and 3-permutable varieties and quasi-varieties of universal algebras.
- Differential involvement of the three nuclear estrogen receptors during oogenesis in European sea bass (Dicentrarchus labrax)Publication . Zapater, Cinta; Molés, Gregorio; Muñoz, Iciar; Pinto, Patricia IS; Canario, Adelino; Gómez, AnaEstrogens are involved in a wide range of processes in vertebrate reproduction through ligand activation of their specific cognate receptors. In most teleosts, three nuclear estrogen receptor subtypes have been identified (Esr1, Esr2a, and Esr2b). Differences in ligand binding affinity and seasonal expression patterns in reproductive tissues among these Esr subtypes suggest distinct roles during oogenesis, vitellogenesis, and spermatogenesis. This study focuses on the role of the Esr subtypes in European sea bass (Dicentrarchus labrax) oogenesis and their endocrine regulation. The coding genes of the three Esr subtypes are highly expressed in reproduction-related tissues such as pituitary, gonad, and liver. Quantification of esr1, esr2a, and esr2b expression in the ovary and liver during a whole reproductive cycle showed different patterns depending on stage and subtype, suggesting differential roles of the three receptors in the regulation of oogenesis and vitellogenesis. Esr2a and Esr2b also showed differences in transcriptional activity and ligand affinity when functionally characterized in HEK293 cells. Finally, for the first time in teleosts, the localization of the three Esr subtypes in ovarian follicles and their regulation by gonadotropins is described. Immunodetection of the receptors revealed different distribution patterns in follicular cells and various subcellular locations of the oocyte. Gonadotropin stimulation of ovarian follicles in different stages of vitellogenesis showed a consistent induction of esrb2b expression by Fsh. All together, these data reinforce the hypothesis that each estrogen receptor plays a specific role in oogenesis.
- A demanda da investigação artística: sobre um espetáculo teatral criado em contexto académicoPublication . Branco, AntónioA institucionalização progressiva das artes nos curricula académicos obriga a pensar as práticas artísticas no contexto da investigação, nomeadamente, do que o artista-investigador terá de acrescentar ao ato de criação para que ele também possa ser considerado ato de investigação. O presente artigo ensaia uma possibilidade de verbalização autorreflexiva relativa a um espetáculo teatral criado em contexto académico, enquanto elemento central de um projeto de mestrado de um estudante que orientei e em que também fui cocriador, na qualidade de encenador e diretor de atores. Nele, reflito sobre questões de investigação artística que estiveram presentes durante o processo de criação, descrevo metodologias adotadas para os resolver e analiso resultados.
- Evaluation of total phenolic content in virgin olive oil using fluorescence excitation-emission spectroscopy coupled with chemometricsPublication . The aim of this study was to explore the feasibility of direct front-face fluorescence measurements coupled with chemometrics for developing multivatiate models for discrimination between virgin olive oils with low and high TPC and determination of TPC concentration. Results Parallel factor analysis and principal component analysis of fluorescence excitation-emission matrices (EEMs) of virgin olive oils revealed different fluorescent properties for samples with low and high TPC. A perfect discrimination of oils with low and high TPC was achieved using partial least squares (PLS) discriminant analysis. The best regression model for the prediction of TPC was based on the PLS analysis of the unfolded entire EEMs (R-2 = 0.951, RPD = 4.0). Conclusions The results show the potential of fluorescence spectroscopy for direct screening of virgin olive oils for TPC. This may contribute to the development of fast screening methods for TPC assessment, providing an alternative to conventional assays. - Open coast seagrass restoration. Can we do it? Large scale seagrass transplantsPublication . Paulo, Diogo; Cunha, Alexandra H.; Boavida, Joana; Serrao, Ester; Gonçalves, Emanuel J.; Fonseca, MarkSome of the major challenges in seagrass restoration on exposed open coasts are the choice of transplant design that is optimal for coastlines periodically exposed to high water motion, and understanding the survival and dynamics of the transplanted areas on a long time-scale over many years. To contribute to a better understanding of these challenges, we describe here part of a large-scale seagrass restoration program conducted in a Marine Park in Portugal. The goal of this study was to infer if it was possible to recover seagrass habitat in this region, in order to restore its ecosystem functions. To infer which methods would produce better long term persistence to recover seagrass habitat, three factors were assessed: donor seagrass species, transplant season, source location. Monitoring was done three times a year for 8 years, in which areas and densities of the planted units were measured, to assess survival and growth. The best results were obtained with the species Zostera marina transplanted during spring and summer as compared to Zostera noltii and Cymodocea nodosa. Long-term persistence of established (well rooted) transplants was mainly affected by extreme winter storms but there was evidence of fish grazing effects also. Our results indicate that persistence assessments should be done in the long term, as all transplants were successful (survived and grew initially) in the short term, but were not resistant in the long term after a winter with exceptionally strong storms. The interesting observation that only the largest (11 m(2)) transplanted plot of Z marina persisted over a long time, increasing to 103 m(2) in 8 years, overcoming storms and grazing, raised the hypothesis that for a successful shift to a vegetated state it might be necessary to overpass a minimum critical size or tipping point. This hypothesis was therefore tested with replicates from two donor populations and results showed effects of size and donor population, as only the larger planting units (PUs) from one donor population persisted and expanded. It is recommended that in future habitat restoration efforts large PUs are considered.
