A review on the current Status of Numerical Weather Prediction in Portugal 2021: surface–atmosphere interactions

Earth system modelling is currently playing an increasing role in weather forecasting and understanding climate change, however, the operation, deployment and development of numerical Earth system models are extremely demanding in terms of computational resources and human effort. Merging synergies has become a natural process by which national meteorological services assess and contribute to the development of such systems. With the advent of joining synergies at the national level, the second edition of the workshop on Numerical Weather Prediction in Portugal was promoted by the Portuguese Institute for the Sea and Atmosphere, I.P. (IPMA), in cooperation with several Portuguese Universities. The event was hosted by the University of Évora, during the period of 11–12 of November 2021. It was dedicated to surface–atmosphere interactions and allowed the exchange of experiences between experts, students and newcomers. The workshop provided a refreshed overview of ongoing research and development topics in Portugal on surface–atmosphere interaction modelling and its applications and an opportunity to revisit some of the concepts associated with this area of atmospheric sciences. This article reports on the main aspects discussed and offers guidance on the many technical and scientific modelling platforms currently under study.
Tracking prostate carcinogenesis over time through urine proteome profiling in an animal model: an exploratory approach

Prostate cancer (PCa) is one of the most lethal diseases in men, which justifies the search for new diagnostic tools. The aim of the present study was to gain new insights into the progression of prostate carcinogenesis by analyzing the urine proteome. To this end, urine from healthy animals and animals with prostate adenocarcinoma was analyzed at two time points: 27 and 54 weeks. After 54 weeks, the incidence of pre-neoplastic and neoplastic lesions in the PCa animals was 100%. GeLC-MS/MS and subsequent bioinformatics analyses revealed several proteins involved in prostate carcinogenesis. Increased levels of retinol-binding protein 4 and decreased levels of cadherin-2 appear to be characteristic of early stages of the disease, whereas increased levels of enolase-1 and T-kininogen 2 and decreased levels of isocitrate dehydrogenase 2 describe more advanced stages. With increasing age, urinary levels of clusterin and corticosteroid-binding globulin increased and neprilysin levels decreased, all of which appear to play a role in prostate hyperplasia or carcinogenesis. The present exploratory analysis can be considered as a starting point for studies targeting specific human urine proteins for early detection of age-related maladaptive changes in the prostate that may lead to cancer.
Pulmonary and gastrointestinal parasitic infections in small ruminant autochthonous breeds from centre region of Portugal—a cross sectional study

The production of small ruminant autochthonous breeds in the Centre region of Portugal is practiced in a semi-extensive husbandry system, exposing animals to parasitic infections. The main objective of this study was to estimate the prevalence of lungworm infection and identify risk factors. Fecal samples of 203 goats and 208 sheep from 30 herds were collected per rectum and subjected to the modified Baermann test. The overall prevalence of infection was 57.7%, significantly higher in goats (95.6%) than in sheep (20.7%) (p < 0.001). According to the binary logistic regression model, sheep dewormed with albendazole, mebendazole plus closantel, or ivermectin plus clorsulon presented a risk of Protostrongylidae infection 29.702, 7.426, or 8.720 times higher, respectively, than those dewormed with eprinomectin. Additionally, the presence of gastrointestinal parasites was investigated in 307 fecal samples using Mini-FLOTAC (R). The overall prevalence of infection was 86.3%, also significantly higher in goats (93.2%) than in sheep (79.9%) (p < 0.001). Strongyle-type eggs were the most frequently identified, both in sheep (69.8%) and goats (87.8%), followed by Eimeria oocysts (40.3% in sheep and 68.9% in goats). Considering the high prevalence and the burden of lungworm parasitic infection, it is urgent to determine its economic impact and the repercussions in animal health in the Centre region of Portugal to establish appropriate therapeutic guidelines.
Antigenotoxic properties of the halophyte Polygonum maritimum L. highlight its potential to mitigate oxidative stress-related damage

Long-term exposure to dietary xenobiotics can induce oxidative stress in the gastrointestinal tract, possibly causing DNA damage and contributing to the initiation of carcinogenesis. Halophytes are exposed to constant abiotic stresses, which are believed to promote the accumulation of antioxidant metabolites like polyphenols. The aim of this study was to evaluate the antioxidant and antigenotoxic properties of the ethanol extract of the aerial part of the halophyte Polygonum maritimum L. (PME), which can represent a dietary source of bioactive compounds with potential to attenuate oxidative stress-related damage. The PME exhibited a high antioxidant potential, revealed by the in vitro capacity to scavenge the free radical DPPH (IC50 = 2.29 +/- 0.10 mu g/mL) and the improved viability of the yeast Saccharomyces cerevisiae under oxidative stress (p < 0.001, 10 min). An antigenotoxic effect of PME against H2O2-induced oxidative stress was found in S. cerevisiae (p < 0.05) with the dominant deletion assay. In vitro colorimetric assays and LC-DAD-ESI/MSn analysis showed that PME is a polyphenol-rich extract composed of catechin, (epi)catechin dimer and trimers, quercetin and myricetin glycosides. Hence, P. maritimum is a source of antioxidant and antigenotoxic metabolites for application in industries that develop products to provide health benefits.
Protective properties of the edible halophyte Carpobrotus edulis (L.) N.E.Br. towards neoformed food contaminants-related oxidative stress and genotoxicity

Carpobrotus edulis (L.) N.E.Br. is an edible halophyte native to South Africa, where it has been traditionally consumed as food, revealing its nutritional value, and used in the treatment of human diseases probably due to the accumulation of bioactive compounds such as polyphenols. These compounds have potential to display valuable biological activities that could bring several benefits to the human being, including the mitigation of toxicity induced by food contaminants. Therefore, this study investigated biological activities with healthpromoting potential of a C. edulis ethanol extract (CEE), such as antioxidant, antigenotoxic, and antimutagenic. CEE was shown by LC-DAD-ESI/MSn to be composed of 16 polyphenols. The extract exhibited antioxidant properties by protecting Saccharomyces cerevisiae viability and improving the redox state of HepG2 cells under oxidative stress. Antigenotoxicity of CEE against H2O2-induced oxidative damage was detected in HepG2 and Caco-2 cells by comet assay and subsequently observed in the dominant deletion assay in S. cerevisiae. Antigenotoxicity and antimutagenicity of CEE towards the neoformed contaminant benzo[a]pyrene was found in HepG2 and Caco-2 cells by comet assay and in Salmonella typhimurium using the Ames test, respectively. The disclosed antigenotoxic and antimutagenic properties of C. edulis further advance its potential for application in food and nutraceutical industries for health benefits.
