- Implicit structured sequence learning: an fMRI study of the structural mere-exposure effectPublication . Folia, Vasiliki; Petersson, Karl MagnusIn this event-related fMRI study we investigated the effect of 5 days of implicit acquisition on preference classification by means of an artificial grammar learning (AGL) paradigm based on the structural mere-exposure effect and preference classification using a simple right-linear unification grammar. This allowed us to investigate implicit AGL in a proper learning design by including baseline measurements prior to grammar exposure. After 5 days of implicit acquisition, the fMRI results showed activations in a network of brain regions including the inferior frontal (centered on BA 44/45) and the medial prefrontal regions (centered on BA 8/32). Importantly, and central to this study, the inclusion of a naive preference fMRI baseline measurement allowed us to conclude that these fMRI findings were the intrinsic outcomes of the learning process itself and not a reflection of a preexisting functionality recruited during classification, independent of acquisition. Support for the implicit nature of the knowledge utilized during preference classification on day 5 come from the fact that the basal ganglia, associated with implicit procedural learning, were activated during classification, while the medial temporal lobe system, associated with explicit declarative memory, was consistently deactivated. Thus, preference classification in combination with structural mere-exposure can be used to investigate structural sequence processing (syntax) in unsupervised AGL paradigms with proper learning designs.
- Cetuximab plus platinum-based chemotherapy in head and neck Squamous Cell Carcinoma: a retrospective study in a single Comprehensive European Cancer InstitutionPublication . De Mello, Ramon Andrade; Gerós, Sandra; Alves, Marcos Pantarotto; Moreira, Filipa; Avezedo, Isabel; Dinis, JoseBackground: The use of cetuximab in combination with platinum (P) plus 5-fluorouracil (F) has previously been demonstrated to be effective in the treatment of metastatic squamous cell cancer of head and neck (SCCHN). We investigated the efficacy and outcome of this protocol as a first-line treatment for patients with recurrent or metastatic disease. We evaluated overall-survival (OS), progression-free-survival (PFS), overall response rate (ORR) and the treatment toxicity profile in a retrospective cohort. Patients and Methods: This study enrolled 121 patients with untreated recurrent or metastatic SCCHN. The patients received PF+ cetuximab every 3 weeks for a maximum of 6 cycles. Patients with stable disease who received PF+ cetuximab continued to receive cetuximab until disease progressed or unacceptable toxic effects were experienced, whichever occurred first. Results: The median patient age was 53 (37-78) years. The patient cohort was 86.8% male. The addition of cetuximab to PF in the recurrent or metastatic setting provided an OS of 11 months (Confidential Interval, CI, 95%, 8.684-13.316) and PFS of 8 months (CI 95%, 6.051-9.949). The disease control rate was 48.9%, and the ORR was 23.91%. The most common grade 3 or 4 adverse events in the PF+ cetuximab regimen were febrile neutropenia (5.7%), skin rash (3.8%) and mucosistis (3.8%). Conclusions: The results of this study suggest that cetuximab plus platinum-fluorouracil chemotherapy is a good option for systemic treatment in advanced SSCHN patients. This regimen has a well-tolerated toxicity profile.
- Assessing the morphological variability of unio delphinus spengler, 1783 (Bivalvia: Unionidae) using geometric morphometryPublication . Morais, Pedro; Rufino, Marta M.; Reis, Joaquim; Dias, Ester; Sousa, RonaldoThe morphological variability of freshwater bivalve species, observed between and within river basins, may hamper their correct identification, even by experienced researchers. Classic morphometric measurements, i.e. shell length, height and thickness, or their ratios, are generally insufficient to distinguish populations and/or species. These issues may be overcome using a geometric morphometric method, which allows analysis of the overall shape of the individual, independently of its size. Thus, we aimed to test the usefulness of two geometric morphometric tools, landmarks and sliding semilandmarks, to evaluate the morphological variability of Unio delphinus Spengler, 1783 in three habitats of the Guadiana Basin (SW Iberian Peninsula, Europe): estuary, river and stream. We used 13 landmarks located on the shell interior (at the teeth, muscle scars and pallial line) and 35 sliding semilandmarks for the shell contour. These morphometric analyses showed that the shell shape of U. delphinus differs significantly among different habitats. Estuarine and stream shells are the most disparate (James index = 649.114, permutation P-value <0.001) and variability is not related to variations in shell size. The main differences in shell morphology are the following: (1) estuarine shells are more elongate, while riverine shells are more subovate; (2) the anterior curvature at the umbo is steeper in estuarine and riverine shells; (3) estuarine shells have an arched curvature at the ventral part of the shell, which is absent in specimens from the other habitats. Our data suggest that the morphology of U. delphinus shells might be influenced by the water flow characteristics of each habitat, since shells exhibited characteristics that are typically observed in freshwater mussels from lotic and lentic habitats.
- Field study on the accumulation of trace elements by vegetables produced in the vicinity of abandoned pyrite minesPublication . Alvarenga, Paula; Simoes, Isabel; Palma, Patrícia; Amaral, Olga; Matos, Joao XavierTo evaluate the accumulation of trace elements (TE) by vegetables produced in the vicinity of abandoned pyrite mines, eighteen different small farms were selected near three mines from the Portuguese sector of the Iberian Pyrite Belt (Sao Domingos, Aljustrel and Lousal). Total and bioavailable As, Cu, Pb, and Zn concentrations were analyzed in the soils, and the same TE were analyzed in three different vegetables, lettuce (Lactuca sativa), coriander (Coriandrum sativum), and cabbage (Brassica oleracea), collected at the same locations. The soils were contaminated with As, Cu, Pb, and Zn, since their total concentrations exceeded the considered soil quality guideline values for plant production in the majority of the sampling sites. The maximum total concentrations for those TE were extremely high in some of the sampling sites (e.g. 1851 mg As kg(-1) in Sao Domingos, 1126 mg Cu kg(-1) in Aljustrel, 4946 mg Pb kg(-1) in Sao Domingos, and 1224 mg Zn kg(-1) in Aljustrel). However, the soils were mainly circumneutral, a factor that contributes to their low bioavailable fractions. As a result, generally, the plants contained levels of these elements characteristic of uncontaminated plants, and accumulation factors for all elements <1, typical of excluder plants. Furthermore, the estimated daily intake (EDI) for Cu and Zn, through the consumption of these vegetables, falls below the recommended upper limit for daily intake of these elements. The sampling site that stood out from the others was located at Sao Joao de Negrilhos (Aljustrel), where bioavailable Zn levels were higher, a consequence of the slight acidity of the soil. Therefore, the Zn content in vegetables was also higher, characteristic of contaminated plants, emphasizing the risk of Zn entering the human food chain via the consumption of crops produced on those soils. - Impacts of CO2-induced seawater acidification on coastal Mediterranean bivalves and interactions with other climatic stressorsPublication . T.; Dellali, M.; Chícharo, LuísThe effects of seawater acidification caused by increasing concentrations of atmospheric carbon dioxide (CO2), combined with other climatic stressors, were studied on 3 coastal Mediterranean bivalve species: the mussel Mytilus galloprovincialis and the clams Chamelea gallina and Ruditapes decussatus. CO2 perturbation experiments produced contrasting responses on growth and calcification of juvenile shells, according to species and location. In the Northern Adriatic (Italy), long-term exposure to reduced pH severely damaged the shells of M. galloprovincialis and C. gallina and reduced growth for the latter species. Seawater in the Ria Formosa lagoon (Portugal) was consistently saturated in carbonates, which buffered the impacts on calcification and growth. After 80 days, no shell damage was observed in Portugal, but mussels in the acidified treatments were less calcified. Reduced clearance, ingestion and respiration rates and increased ammonia excretion were observed for R. decussatus under reduced pH. Clearance rates of juvenile mussels were significantly reduced by acidification in Italy, but not in Portugal. Both locations showed a consistent trend for increased ammonia excretion with decreasing pH, uggesting increased protein catabolism. Respiratory rates were generally not affected. Short-term factorial experiments done in Italy revealed that acidification caused alterations in immunological parameters of adult bivalves, particularly at temperature and salinity values far from the optimal for the species in the Mediterranean. Overall, our results showed large variations in the sensitivities of bivalves to climatic changes, among different species and between local populations of the same species. Expectations of impacts, mitigation and adaptation strategies have to consider such local variability.
- The freshwater Sponge Ephydatia Fluviatilis harbours diverse pseudomonas species (Gammaproteobacteria, Pseudomonadales) with broad-spectrum antimicrobial activityPublication . Pseudomonas species (Gammaproteobacteria, Pseudomonadales) are capable of colonizing a broad range of eukaryotic hosts, but knowledge of their diversity and function in freshwater invertebrates is rudimentary. We assessed the diversity, structure and antimicrobial activities of Pseudomonas spp. in the freshwater sponge Ephydatia fluviatilis. Polymerase Chain Reaction - Denaturing Gradient Gel Electrophoresis (PCR-DGGE) fingerprints of the global regulator gene gacA revealed distinct structures between sponge-associated and free-living Pseudomonas communities, unveiling previously unsuspected diversity of these assemblages in freshwater. Community structures varied across E. fluviatilis specimens, yet specific gacA phylotypes could be detected by PCR-DGGE in almost all sponge individuals sampled over two consecutive years. By means of whole-genome fingerprinting, 39 distinct genotypes were found within 90 fluorescent Pseudomonas isolates retrieved from E. fluviatilis. High frequency of in vitro antibacterial (49%), antiprotozoan (35%) and anti-oomycetal (32%) activities was found among these isolates, contrasting less-pronounced basidiomycetal (17%) and ascomycetal (8%) antagonism. Culture extracts of highly predation-resistant isolates rapidly caused complete immobility or lysis of cells of the protozoan Colpoda steinii. Isolates tentatively identified as P. jessenii, P. protegens and P. oryzihabitans showed conspicuous inhibitory traits and correspondence with dominant sponge-associated phylotypes registered by cultivation-independent analysis. Our findings suggest that E. fluviatilis hosts both transient and persistent Pseudomonas symbionts displaying antimicrobial activities of potential ecological and biotechnological value.
- Cytogenetic and histological studies of the brook trout, Salvelinus fontinalis (Mitchill), and the Arctic char, S-alpinus (L.) hybridsPublication . Ocalewicz, Konrad; Hliwa, Piotr; Pomianowski, Konrad; Lisboa, Rodrigo; Jankun, MalgorzataAlthough brook trout and the Arctic char hybrids are able to reproduce, individuals with decreased fertility or even fish that are unable to produce any gametes have been also described. Abnormal gonadal development and disturbances in the gamete production in the char hybrid offspring may be triggered by the odd chromosome number and disturbances in their pairing during meiosis. To verify this hypothesis, cytogenetic examination and the gonadal histology analysis of the brook trout x Arctic char hybrids were carried out. Diploid chromosome number in the studied char (F-1) hybrids varied from 82 to 84 (FN = 99-102). Among 28 hybrids, 12 males, three females, nine intersex individuals and two sterile specimens were described. In the case of two individuals, gonads were not found. Diploid chromosome numbers in the males and intersex individuals varied from 82 to 84. Chromosome numbers in the females were 82 and 83 chromosomes. Two sterile fish exhibited karyotypes composed of 82 and 84 chromosomes. Predominance of the ovarian component in the intersex gonads and gonadal sex ratio distortion towards the males suggested hybrid females had problems with gonadal differentiation. However, the lack of the clear relationship between chromosome number and gonadal development in the studied hybrids did not support our hypothesis that odd chromosome number may be responsible for such reproductive disturbances in the hybrid individuals. We have presumed that sterility and intersexual development of the gonads may be caused by interactions between brook trout and Arctic char genes on the sex chromosomes and autosomes rather than unpairing of the parental chromosomes.
- Mir-20a regulates in vitro mineralization and BMP signaling pathway by targeting BMP-2 transcript in fishPublication . Tiago, Daniel; Marques, C. L.; Roberto, Vania Palma; Cancela, Leonor; Laizé, VincentMicroRNAs (miRNAs) are important regulators of vertebrate development but their role during skeletogenesis remains unknown. In this regard, we investigated the mineralogenic activity of miR-20a, a miRNA associated with osteogenesis, in fish bone-derived cells. Expression of miR-20a was up-regulated during differentiation and its overexpression inhibited mineralization, suggesting a role in fish tissue calcification. In this regard, a conserved miR-20a binding site was identified in bone morphogenetic protein 2 (BMP-2) 30UTR and its functionality was evidenced through luciferase assays, and further confirmed by western-blot and qPCR. Type II BMP receptor (BMPR2) is also targeted by miR-20a in mammalian systems and evidence was collected for the presence of a binding site in fish sequences. We propose that miR-20a is a regulator of BMP pathway through specific action on BMP-2 and possibly BMPR2. Overexpression of miR-20a was also shown to up-regulate matrix Gla protein (MGP) transcript, a physiological inhibitor of calcification previously found to form a complex with BMP-2. We propose that MGP may play a role in the anti-mineralogenic effect promoted by miR-20a by decreasing availability of BMP-2. This study gives new insights into miRNA-mediated regulation of BMP-2, and sheds light into the potential role of miR-20a as a regulator of skeletogenesis.
- Teorias leigas sobre saúde e doença e conceitos relacionados em pessoas idosas.Publication . Luísa, Cláudia; Pérez, FranciscoO envelhecimento humano é um processo irreversível e contínuo, que acarreta diversas mudanças a nível físico, social, cognitivo, psicológico, entre outros. Neste sentido é necessário e urgente prevenirmos e educarmos a população em geral e os idosos em particular, para um envelhecimento bem-sucedido. Assim, no sentido de conhecermos um pouco melhor o que pensam e sentem os idosos, realizou-se a investigação “Teorias leigas sobre saúde e doença e conceitos relacionados em pessoas idosas”, a uma amostra de 300 idosos, de diferentes contextos (universidades da terceira idade, associações de desenvolvimento local, e também em idosos sem qualquer ligação institucional), na região Algarvia. Os objetivos gerais da investigação passaram pela compreensão dos significados leigos de conceitos como: saúde, doença, qualidade de vida, felicidade, satisfação com a vida, otimismo e stress em adultos com idade igual ou superior a 60 anos, bem como por encontrar possíveis variáveis relevantes que podem ajudar a explicar as características de cada grupo e encontrar possíveis relações existentes entre as variáveis sociodemográficas e os diversos aspetos psicológicos em estudo. Para a recolha de dados foi administrado um protocolo que continha um questionário de dados demográficos, uma entrevista semiestruturada, com algumas questões sobre teorias leigas de saúde, doença, qualidade de vida, felicidade, satisfação vital, otimismo e stress, uma escala de avaliação da qualidade de vida (Hawthorne, Richardson, Osborne, & McNeil, 1997) uma de satisfação com a vida (Diener, Emmons, Larsen, & Griffin, 1985), uma de espiritualidade (Pais-Ribeiro & Pinto, 1997) e uma escala de orientação vital (Carver & Bridges, 1994). Os resultados obtidos permitiram atingir os objetivos inicialmente traçados e chegar a diversas conclusões. Assim, foi possível identificar as categorias que melhor caracterizam os conceitos leigos de saúde, doença, qualidade de vida, felicidade, satisfação com a vida, otimismo e stress, que são diferentes para cada tema/dimensão e questão, quer seja na perspetiva individual ou grupal. Identificou-se 179 categorias, embora somente 26 delas se revelassem significativas (com uma expressividade igual ou superior a 30% e nas situações em que esse número não foi atingido, foi tido em conta o valor mais elevado), e com essas realizou-se todas as outras análises. As categorias mais mencionadas foram a comportamental (sete vezes), saúde (cinco vezes) e psicológica (quatro vezes), atendendo aos temas em análise. Ao se aplicar o método da análise discriminante e da regressão logística foi possível identificar as variáveis preditoras, que melhor explicam os temas para esta amostra extraída de uma população de pessoas idosas. De igual forma, foi possível conhecer as perceções dos idosos acerca dos diversos constructos, sendo que de uma maneira geral, percecionam-se saudáveis, pouco doentes, com uma qualidade de vida, felicidade e satisfação com a vida moderadas. Em relação às expectativas futuras, os dados confirmam que a maioria deles é otimista e que se apresenta pouco stressada. Estes dados também foram corroborados com os resultados provenientes dos instrumentos de qualidade de vida, satisfação com a vida, espiritualidade e orientação vital, dado que os valores médios encontrados foram moderados e, em alguns casos, elevados.
- Characterization of polymorphic microsatellite loci in the Antarctic krill Euphausia superbaPublication . Candeias, Rui; Teixeira, Sara; Duarte, C. M.; Pearson, G. A.Background: The Antarctic krill, Euphausia superba is a pelagic crustacean, abundant in high-density swarms (10 000 – 30 000 ind/m2) with a circumpolar distribution and a key role in the food web of the Southern Ocean. Only three EST derived microsatellite markers have been used in previous genetic studies, hence we developed additional highly polymorphic microsatellite markers to allow robust studies of the genetic variability and population differentiation within this species. Findings: The microsatellite markers described here were obtained through an enriched genomic library, followed by 454 pyrosequencing. A total of 10 microsatellite markers were tested in 32 individuals from the Antarctic Peninsula. One of the tested loci was fixed for one allele while the other was variable. Of the remaining nine markers, seven showed no departure from Hardy-Weinberg equilibrium. The mean number of alleles was 14.9. Conclusions: These markers open perspectives for population genetic studies of this species to unravel genetic structure, dispersal and population biology, vital information for future conservation.
