However, for a safe use of reprogrammed cells in new clinical applications, the reprogramming process remains to be improved. We have showed that the use of the protein Cited2 in during the reprogramming procedure increases the efficiency and homogenizes the process.
- Field measurements and hydrodynamic modelling to evaluate the importance of factors controlling overwashPublication . Matias, Ana; Carrasco, A.R.; Loureiro, Carlos; Masselink, Gerd; Andriolo, Umberto; McCall, Robert; Ferreira, Oscar; Plomaritis, Theocharis; Pacheco, André; Guerreiro, MarthaOverwash hydrodynamic datasets are mixed in quality and scope, being difficult to obtain due to fieldwork experimental limitations. Nevertheless, these measurements are crucial to develop reliable models to predict overwash. Aiming to overcome such limitations, this work presents accurate fieldwork data on overwash hydrodynamics, further exploring it to model overwash on a low-lying barrier island. Fieldwork was undertaken on Barreta Island (Portugal) in December 2013, during neap tides and under energetic conditions, with significant wave height reaching 2.6 m. During approximately 4 h, more than 120 shallow overwash events were measured with a video-camera, a pressure transducer and a current-meter. This high-frequency fieldwork dataset includes runup, overwash number, depth and velocity. Fieldwork data along with information from literature were used to implement XBeach model in non-hydrostatic mode (wave-resolving). The baseline model was tested for six verification cases; and the model was able to predict overwash in five. Based in performance metrics and the verification cases, it was considered that the Barreta baseline overwash model is a reliable tool for the prediction of overwash hydrodynamics. The baseline model was then forced to simulate overwash under different hydrodynamic conditions (waves and lagoon water level) and morpho-sedimentary settings (nearshore topography and beach grain-size), within the characteristic range of values for the study area. According to the results, the order of importance of factors controlling overwash predictability in the study area are: 1st) wave height (more than wave period) can promote overwash 3–4 times more intense than the one recorded during fieldwork; 2nd) nearshore bathymetry, particularly shallow submerged bars, can promote an average decrease of about 30% in overwash; 3rd) grain-size, finer sediment produced an 11% increase in overwash due to reduced infiltration; and 4th) lagoon water level, only negligible differences were evidenced by changes in the lagoon level. This implies that for model predictions to be reliable, accurate wave forecasts are necessary and topo-bathymetric configuration needs to be monitored frequently.
- Warfarin-exposed zebrafish embryos resembles human warfarin embryopathy in a dose and developmental-time dependent manner - From molecular mechanisms to environmental concernsPublication . Since it crosses placental barrier it can induce warfarin embryopathy (WE), a fetal mortality in neonates characterized by skeletal deformities in addition to brain hemorrhages. Although the effects of warfarin exposure in aquatic off target species were already described, the particular molecular toxicological mechanisms during early development are still unclear. Here, we used zebrafish (Danio rerio) to describe and compare the developmental effects of warfarin exposure (0, 15.13, 75.68 and 378.43 mM) on two distinct early developmental phases (embryos and eleuthero-embryos). Although exposure to both developmental phases induced fish mortality, only embryos exposed to the highest warfarin level exhibited features mimicking mammalian WE, e.g. high mortality, higher incidence of hemorrhages and altered skeletal development, among other effects. To gain insights into the toxic mechanisms underlying warfarin exposure, the transcriptome of embryos exposed to warfarin was explored through RNA-Seq and compared to that of control embryos. 766 differentially expressed (564 up- and 202 down-regulated) genes were identified. Gene Ontology analysis revealed particular cellular components (cytoplasm, extracellular matrix, lysosome and vacuole), biological processes (mainly amino acid and lipid metabolism and response to stimulus) and pathways (oxidative stress response and apoptosis signaling pathways) being significantly over-represented in zebrafish embryos upon warfarin exposure. Protein-protein interaction further evidenced an altered redox system, blood coagulation and vasculogenesis, visual phototransduction and collagen formation upon warfarin exposure. The present study not only describes for the first time the WE in zebrafish, it provides new insights for a better risk assessment, and highlights the need for programming the rat eradication actions outside the fish spawning season to avoid an impact on off target fish community. The urge for the development of more species-specific anticoagulants for rodent pest control is also highlighted.
- A preliminary study on ochre sources in Southwestern Germany and its potential for ochre provenance during the Upper PaleolithicPublication . Velliky, Elizabeth C.; Barbieri, Alvise; Porr, Martin; Conard, Nicholas J.; MacDonald, Brandi LeeThe use of mineral pigments, specifically iron-oxide rich mineral pigments called ochre, has been put forward as a key element in the development of symbolic and non-utilitarian behaviors in human evolution. However, the processes of ochre procurement, trade and use are difficult to conceptualize without the identification and characterization of the sources where these materials were acquired. We present the results of geochemical analyses of ochre source samples collected from the Swabian Jura, Black Forest, and other localities in southern and eastern Germany. The goal of this study was to build the groundwork for future investigations on the range of ochre behaviors at archaeological sites in the region. We aimed to determine whether specific ochre outcrops could be differentiated based on their geochemical signatures. Using data from Neutron Activation Analysis (NAA), we were able to determine that the ochre source regions exhibit greater source inter-variability than infra-variability when observed using a range of statistical techniques, therefore satisfying the provenance postulate. Furthermore, the data provide the foundation for a Central European database of ochre sources to allow the comparison of ochres from different regions to archaeological ochres from important nearby and perhaps distant sites.
- Dietary tryptophan supplementation induces a transient immune enhancement of gilthead seabream (Sparus aurata) juveniles fed fishmeal-free dietsPublication . C.; Dias, Jorge; Silva, Tome S.; Costas, BenjaminEuropean aquaculture is an industry with a high sustainability profile contributing to the supply of safe seafood. However, several diseases can affect farmed fish and it is imperative to find alternatives for chemotherapeutic treatments when disease outbreaks occur. Maintenance of health through nutrition is well-establish in modern animal farming, and amino acids (AA) are promising candidates as functional additives to improve fish health. Therefore, the goal of this research is to provide a better understanding of the influence of tryptophan supplementation on nutritional condition and immune mechanisms in fish. Triplicate groups of fish (13.3 +/- 0.3g) previously fed with a fishmeal-based diet were either fed a control diet with an extreme formulation (0% fishmeal) but meeting the AA requirements (CTRL), or the SUP diet, formulated as the CTRL with an increase in tryptophan (TRP) content. After 2 and 13 weeks of feeding, head-kidney (HK), liver (L) and white skeletal muscle (WSM) were collected for gene expression, whereas plasma was suited for humoral immune parameters. A holistic approach using transcriptomic, humoral and zootechnical parameters was undertaken. The expression of 29-31 genes for WSM, L or HK confirms an effect due to the treatment across time. A two-way ANOVA analysis revealed that 15-24 genes varied significantly depending on the tissue, and the multivariate analysis by means of PLS-DA explained (R-2) and predicted (Q(2)) with four components up to 93% and 78% of total variance, respectively. Component 1 (R-2 = 50.06%) represented the time effects, whereas components 2 (24.36%) and 3 (13.89%) grouped fish on the basis of dietary treatment, at early sampling. The HK results in particular suggest that fish fed SUP diet displayed an immunostimulated state at 2 weeks. No major differences were observed in plasma humoral parameters, despite an increase in antiprotease and peroxidase activities after 13 weeks regardless of dietary treatment. These results suggest that tryptophan supplementation may improve the seabream immune status after 2 weeks. Hence, the use of functional feeds is especially relevant during a short-term feeding period before a predictable stressful event or disease outbreak, considering that these putative advantageous effects seem to disappear after a 13 weeks feeding period.
- Modelling wine tourism experiencesPublication . Madeira, Arlindo; Correia, Antónia; Filipe, Jose AntonioWine experiences contributed to attracting tourists to the destinations, in particular in Portugal where wine is worldwide renowned. This research aims to understand what drives tourists to visit a destination to live a wine experience and how this experience may improve the image and retain tourists. This model conceptual in its essence was applied in an emergent wine region in Portugal - Lisbon. Data were collected in 16 wineries from a random sample of 314 visitors in an emergent wine region, and structural equation modelling (SEM) was applied. Structural equation modelling was used to test hypotheses and validate the conceptual model. Results show that motivation positively influences the wine experiences, which in turn is explained by the dimensions presented in the study. Furthermore, the wine experience influences image and satisfaction positively and the brand equity negatively. From the managerial perspective, implications are discussed as well as the main limitations, and then suggestions for further research are provided.
- Understanding the individual role of fish, oyster, phytoplankton and macroalgae in the ecology of integrated production in earthen pondsPublication . Cunha, M. E.; Quental-Ferreira, H.; Parejo, A.; Gamito, Sofia; Ribeiro, L.; Moreira, Márcio; Monteiro, I; Soares, F.; Pousao-Ferreira, P.In order to demonstrate that IMTA can be profitable and a good alternative to regular semi-intensive fish mariculture production in earthen ponds three different production treatments with distinct combinations of trophic levels were designed: (i) a combination of fish, filter feeders, phytoplankton and macroalgae, (ii) fish, filter feeders and phytoplankton and (iii) fish, phytoplankton and macroalgae, to evaluate the role of each trophic level within an Integrated Multi-Trophic Aquaculture system (IMTA). Each treatment was carried out under semi-intensive conditions with two replicates, in a total of 6 earthen ponds of 500 m(2) surface and depth of 1.5 m. The results showed that fish, oyster, phytoplankton and macroalgae integrated aquaculture is a healthy sustainable production system for mariculture in earthen ponds, providing much more fish supply compared with the other two treatments. Ponds with filter feeders had significantly lower turbidity (Nephelometric Formazin unit (FNU) of 13 in the morning and 17 in the afternoon) when compared to ponds without filter feeders (16 FNU in the morning and 20 FNU in the afternoon) with increased light penetration throughout water column (61 and 55 cm transparency in ponds with filter feeders compared to 51 cm in ponds without filter feeders) and consequently higher photosynthetic activity with significantly higher dissolved oxygen (5.4 mg L in the morning and 6.7 mg L-1 in the afternoon in ponds with filter feeders compared to 5.3 mg L-1 in the morning and 6.4 mg L-1 in the afternoon in ponds without filter feeders) and carbon sequestration (0.50 and 0.53 mg L-1 8 h(-1) in ponds with filter feeders and 0.43 mg L-1 8 h(-1) in ponds without filter feeders). In the fish, filter feeder, phytoplanton and macroalgae IMTA treatment, phytoplankton played a crucial role because they increased DO levels, removed the excess of nutrients from animal excretion, and was used as food by the filter feeders. Almost as important is the presence of filter feeders since they control the density of the microalgae and particulate matter in the ponds contributing to a more constant level of DO and higher transparency of the water column. The increased transparency and pond fertilization by oyster excretion, resulted in higher proliferation of phytoplankton (chlorophyll a concentrations of 16.5 mu gL(-1) and 20.2 mu g L-1 in ponds with filter feeder and 13.3 mu g L-1 in ponds without filter feeder) with benefits not only for filter feeders themselves but also for the macroalgae. At the end there was higher water quality and higher savings (14% day(-1)) in the energy costs for pond aeration. Meagre, white seabream and flathead grey mullet enhance their performance in IMTA systems with the presence of filter feeders with food conversion rates (FCR) of 1.52 when compared with 2.46 in the regular semi-intensive system composed by fish, phytoplankton and macroalgae. Meagre grew significantly more in IMTA systems with controlled macroalgae while white seabream and flathead grey mullet enhance their performance in the presence of macroalgae. The results show that the fish, oyster, phytoplankton and macroalgae integrated production in earthen ponds is an improved system compared to the regular semi-intensive fish production. The enhanced water quality in these systems leads to improved fish performance and higher biomass production, and to reduction in the energy power used, contributing to greater profitability.
- The role of residents' apathy in tourism: a tourist perspectivePublication . To test and conceptualize residents' apathy, this study aims to filling a gap by proposing a theoretical model and testing it in three different destinations, namely, Lisbon (Portugal), Isfahan (Iran) and Olbia (Italy). Three convenience samples were used for the purposes. Structural equation modelling (SEM) was used to test and validate the model. This study contributes to the current body of knowledge by validating a conceptual model that aims to analyse how residents' apathy (i.e. lack of interest, lack of initiative, alienation and environmental-based apathy), as perceived by tourists, affects the host-guest interaction and perceived service quality and visitors' behavioural intentions. Managerial implications and future research are discussed.

However, the toxicity and reactivity of the spent chromophore, o-nitrosobenzaldehyde, remains an unaddressed difficulty. In this study we have applied the well-established supramolecular photochemical concepts to retain the spent trigger o-nitrosobenzaldehyde within the organic capsule after release of water-soluble acids and alcohols. The sequestering power of organic capsules for spent chromophores during photorelease from ortho-nitrobenzyl esters, ethers and alcohols is demonstrated with several examples.

Jellyfish are gelatinous organisms, with a life cycle composed by sessile stage (polyp) and free living stage (medusa). Blooms jellies, which may have negative impact on fisheries and tourism, are strongly driven by the availability of food, since predators are relatively reduce. The diet of jellies includes small crustacean, egg and fish larvae. Yet, recent UAlg studies showed a much more diverse diet (ex: drifting microbes, terrestrial detritus). Major contributors for ocean jellification are closed connect to men activities and include: (1) excessive input of nutrients (ex: sewage agriculture fertilizers), (2) overfishing of fishes and sea turtles; (3) introduction of artificial structures and (4) changes in hydrological cycle (ex: river dams).
