- Application of multidimensional and conventional fluorescence techniques for classification of beverages originating from various berry fruitPublication . Sikorska, Ewa; Wlodarska, Katarzyna; Khmelinskii, IgorThe objectives of this study were to characterize fluorescence of beverages from berry fruit, including chokeberry, blackcurrant, raspberry and strawberry, and to develop classification models based on different types of fluorescence spectra to identify beverages depending on the fruit species. Total fluorescence spectra (excitation-emission matrices, EEMs) and total synchronous fluorescence spectra (TSFS) were recorded for a series of commercial berry fruit beverages. An analysis of EEMs using parallel factor analysis (PARAFAC) revealed four components characterized by the excitation/emission maxima at 275/326, 319/410, 414/600, and 360/460 nm, respectively. Based on the spectral profiles, these components were assigned to various groups of phenolic compounds. Partial least squares discriminant analysis was used to develop the classification models. The analysis was performed on PARAFAC scores, unfolded EEMs (uEEMs), unfolded TSFS (uTSFS), and additionally on conventional emission spectra (EMS) measured at particular excitation wavelengths and single synchronous fluorescence spectra (SFS). The classification models with the same average classification error of 4.86% were obtained for the analysis of both the entire uEEMs and uTSFS. Among models based on the individual spectra, the lowest error of 4.42% was obtained for SFS measured at Delta lambda = 40 nm, and an error of 7.64% was obtained for EMS measured at the excitation wavelength of 360 nm. The classification model based on the PARAFAC scores had the highest error of 15.27%. The present results show good potential of fluorescence as rapid and reagent-free tool for authenticity evaluation of berry beverages.
- Evaluation of MGP gene expression in colorectal cancerPublication . Caiado, Helena; Conceição, Natércia; Tiago, Daniel; Marreiros, Ana; Vicente, Susana; Enriquez, Jose Luis; Vaz, Ana Margarida; Antunes, Artur; Guerreiro, Horacio; Caldeira, Paulo; Leonor Cancela, M.Purpose: Matrix Gla protein (MGP) is a vitamin K-dependent, gamma-carboxylated protein that was initially found to be a physiological inhibitor of ectopic calcifications affecting mainly cartilage and the vascular system. Mutations in the MGP gene were found to be responsible for a human pathology, the Keutel syndrome, characterized by abnormal calcifications in cartilage, lungs, brain and vascular system. MGP was recently implicated in tumorigenic processes such as angiogenesis and shown to be abnormally regulated in several tumors, including cervical, ovarian, urogenital and breast. This fact has triggered our interest in analyzing the expression of MGP and of its regulator, the transcription factor runt related transcription factor 2 (RUNX2), in colorectal cancer (CRC). Methods: MGP and RUNX2 expression were analyzed in cancer and non-tumor biopsies samples from 33 CRC patients and 9 healthy controls by RT-qPCR. Consequently, statistical analyses were performed to evaluate the clinical-pathological significance of MGP and RUNX2 in CRC. MGP protein was also detected by immunohistochemical analysis. Results: Showed an overall overexpression of MGP in the tumor mucosa of patients at mRNA level when compared to adjacent normal mucosa and healthy control tissues. In addition, analysis of the expression of RUNX2 mRNA demonstrated an overexpression in CRC tissue samples and a positive correlation with MGP expression (Pearson correlation coefficient 0.636; p <= 0.01) in tumor mucosa. However correlations between MGP gene expression and clinical-pathological characteristics, such as gender, age and pathology classification did not provide relevant information that may shed light towards the differences of MGP expression observed between normal and malignant tissue. Conclusions: We were able to associate the high levels of MGP mRNA expression with a worse prognosis and survival rate lower than five years. These results contributed to improve our understanding of the molecular mechanism underlying MGP deregulation in cancer.
- Congruence between fine-scale genetic breaks and dispersal potential in an estuarine seaweed across multiple transition zonesPublication . Nicastro, Katy; Assis, J.; Serrao, Ester; Pearson, Gareth; Neiva, J.; Valero, Myriam; Jacinto, Rita; Zardi, Gerardo, IGenetic structure in biogeographical transition zones can be shaped by several factors including limited dispersal across barriers, admixture following secondary contact, differential selection, and mating incompatibility. A striking example is found in Northwest France and Northwest Spain, where the estuarine seaweed Fucus ceranoides L. exhibits sharp, regional genetic clustering. This pattern has been related to historical population fragmentation and divergence into distinct glacial refugia, followed by post-glacial expansion and secondary contact. The contemporary persistence of sharp ancient genetic breaks between nearby estuaries has been attributed to prior colonization effects (density barriers) but the effect of oceanographic barriers has not been tested. Here, through a combination of mesoscale sampling (15 consecutive populations) and population genetic data (mtIGS) in NW France, we define regional genetic disjunctions similar to those described in NW Iberia. Most importantly, using high resolution dispersal simulations for Brittany and Iberian populations, we provide evidence for a central role of contemporary hydrodynamics in maintaining genetic breaks across these two major biogeographic transition zones. Our findings further show the importance of a comprehensive understanding of oceanographic regimes in hydrodynamically complex coastal regions to explain the maintenance of sharp genetic breaks along continuously populated coastlines.
- Firm preparation for ISO 9001 certification: the case of the hotel industry in PortugalPublication . Esgarrancho, Sandra; Candido, CarlosThe purpose of this study is to assess whether companies conduct preparations, such as managing culture, before starting to implement an ISO 9001 Quality Management System (QMS). Based on a literature review, the paper develops a model of firm preparation for ISO 9001 certification and several research hypotheses. The survey research method adopted consists of data collection through questionnaires – sent to hotels of four and five stars, in Portugal, in two different moments (2012 and 2014) – and of a longitudinal data analysis based on non-parametric statistical tests. Results show that the majority of companies conduct preparations before starting to implement an ISO 9001 QMS. However, most companies do not assess their initial situation, nor do they plan for the preparations. One of the significant implications from this research is that a more coherent and integrated approach in ISO 9001 preparation is required. Other implications for practice and for research are also noted. This is the first study, as far as the authors are aware of, that addresses the topic of planning for the preparations that a company can make before starting to implement an ISO 9001 QMS.
- Volt-ampere characteristics of porcine retinal Muller cell intermediate filamentsPublication . Makarov, Vladimir; Khmelinskii, IgorIn the current study we reported current-voltage (I/V) characteristics of Muller cell (MC) intermediate filaments (IFs) isolated from porcine retina. It was found that the measured I/V dependences demonstrate behavior of a semiconductor in contact with metal (Au) electrodes. The analysis of the temperature dependence of the experimental I/V curves produced estimates of the parameter values characterizing the electrical conductivity properties of the studied MC IFs. The I/V characteristics and the parameter values allowed to describe the MC IFs as a semiconducting material. The observed properties clarify the mechanism of high-contrast daylight vision of vertebrate eyes. This mechanism was extensively discussed earlier, and is directly dependent on the electric conductivity properties of MC IFs. Significance Statement: Retinal cones and rods are physically connected to glial Muller cells by intermediate filaments (IFs). Electric conductivity of IFs allows for a simple quantum mechanical description of light energy transmission through the retina, providing a convincing mechanism that achieves high-contrast vision in vertebrate eyes. Note that classic light transmission is hindered by light scattering on the retinal structures. Electric conductivity of IFs also opens the possibility for energy exchange within and between other cell types by way of IFs and microtubules, and the possibility that such structures may be used for signaling e.g. in the nervous system.
- Urban agriculture as a keystone contribution towards securing sustainable and healthy development for cities in the futurePublication . Skar, S. L. A.; Ćujić, M.; Tzortzakis, N.; Chrysargyris, A.; Peticila, A.; Alencikiene, G.; Monsees, H.; Junge, R.esearch and practice during the last 20 years has shown that urban agriculture can contribute to minimising the effects of climate change by, at the same time, improving quality of life in urban areas. In order to do so most effectively, land use and spatial planning are crucial so as to obtain and maintain a supportive green infrastructure and to secure citizens' healthy living conditions. As people today trend more towards living in green and sustainable city centres that can offer fresh and locally produced food, cities become again places for growing food. The scope of urban agriculture thereby is to establish food production sites within the city's sphere; for example, through building-integrated agriculture including concepts such as aquaponics, indoor agriculture, vertical farming, rooftop production, edible walls, as well as through urban farms, edible landscapes, school gardens and community gardens. Embedded in changing urban food systems, the contribution of urban agriculture to creating sustainable and climate-friendly cities is pivotal as it has the capacity to integrate other resource streams such as water, waste and energy. This article describes some of the current aspects of the circular city debate where urban agriculture is pushing forward the development of material and resource cycling in cities.
- Consensus must be found on intravenous fluid therapy management in trauma patientsPublication . Mota, Mauro Neves; Santos, M. R.; Cunha, M.; Melo, Filipe; Neves, H.; Abrantes, T.Introduction: Trauma is an important cause of death among young people and 30-40% of this mortality rate is due to hypovolemic shock, intensified by trauma's lethal triad: Hypothermia, Acidosis, and Coagulopathy. Nurses are responsible for managing fluid therapy administration in trauma victims. The purpose of this study is to analyse the reasons why intravenous fluid therapy is recommended for trauma patients' hemodynamic stabilization. Methods: This narrative literature review included published and unpublished studies in English, Spanish or Portuguese between 1994 and January 2019. The search results were analyzed by two independent reviewers. Inclusion criteria encompasses quantitative studies involving trauma victims aged over 18 who underwent fluid therapy in a prehospital assessment context. Results&Discussion: 11 quantitative studies were included. 9 involved the use of fluid therapy for hypotension treatment and 2 of the studies analyzed involved the use of warmed fluid therapy for hypothermia treatment. The analysis performed reveals that the administration of aggressive fluid therapy seems to be responsible for the worsening of the lethal triad. In the presence of traumatic brain injury, permissive hypotension is not allowed due to the negative impact on cerebral perfusion pressure. Used as warming measure, warmed fluid therapy does not seem to have a significant impact on body temperature. Conclusions: There is no consensus regarding the administration of fluid therapy to trauma patients. This conclusion clearly supports the need to develop more randomized controlled trials in order to understand the effectiveness of such measure when it comes to control hypovolemia and hypothermia.
- Linking stress coping styles with brain mRNA abundance of selected transcripts for Senegalese sole (Solea senegalensis) juvenilesPublication . Fatsini, Elvira; Rey, Sonia; Ibarra-Zatarain, Zohar; Boltaña, Sebastián; Mackenzie, Simon; Duncan, Neil J.In fish, proactive and reactive individual stress copying styles (SCS) have been used to resolve variation in molecular expression data. Stress coping styles have been previously described in several stages of Solea senegalensis by validating for the species the use of standard behavioural screening tests. The present study aimed to link behavioural SCS tests with brain transcript abundance in early Senegalese sole juveniles in order to observe the natural variation in a molecular pathway in this species. A total of 50 juveniles were subjected to three individual behavioural (Restraining, New environment and Confinement) and one group (Risk-taking) screening tests. The fish were classified in SCS categories by applying a hierarchical cluster to the variable "Total activity" (the total activity time that the fish was moving in each individual test). Three categories were defined, proactive, intermediate and reactive sole. Six transcripts were chosen and tested, one related to basic metabolism (gapdh-2), three to feeding behaviour (per1, igf-Ia, pparß) and two to the stress response (crh-BP and hsp90aa) in 30 juveniles (10 individuals per SCS category) using rt-qPCR to observe differences in the abundance of those transcripts among SCS. Four transcripts were differentially expressed (DETs) among them. The transcript gapdh-2 showed up-regulation for proactive and intermediate SCS sole while reactive individuals showed down-regulation. Target mRNAs per1, igf-Ia and pparß, showed different levels of up-regulation for proactive and reactive fish while intermediates were highly down-regulated. Surprisingly no differences in stress related transcripts were observed. Correlations were found between variation in coping styles and variation in the abundance of mRNAs involved in important biological functions in Senegalese sole. These results are the first evidence of the relationship between the behavioural individual variation and the fluctuation in brain transcripts abundance in Senegalese sole.
- RELOAD/CoAP architecture for the federation of wireless sensor networksPublication . Pisco, Luís; Guerreiro, Joel; Correia, NoéliaSensing devices are expected to interconnect over large geographical areas and federations of wireless sensor networks are expected in a near future. In such environments a critical issue is how to discover the resources available at devices in a scalable manner. For this purpose, a Constrained Application Protocol (CoAP) Usage for REsource LOcation And Discovery (RELOAD), a generic self-organizing Peer-to-Peer (P2P) overlay network service, has been defined to be used as a lookup service, to store available device resources and as a cache for sensor data. Each P2P resource, at the RELOAD/CoAP overlay, includes references to device resources, hosted at one or more constrained nodes, but no provision is made for the insertion of bindings/references to P2P resources already available at the P2P overlay network. Such feature would increase the efficiency and consistency of storage, avoiding duplicate references, a very relevant issue for future IoT applications relying on the federation of wireless sensor networks. In this article an extension to the service provided by CoAP Usage is proposed so that resource bindings can be managed. Two binding models, and a heuristic algorithm for their implementation, are proposed. Results show that such models lead to a better resource organization, reducing the number of sensor resource entries and/or fetches to the P2P overlay.
- Baseline assessment of underwater noise in the Ria FormosaPublication . Soares, C; Pacheco, André; Zabel, Friedrich; G-Gorbeña, Eduardo; Sequeira, ClaudiaThe Ria Formosa is a sheltered large coastal lagoon located on the Atlantic South Coast of Portugal, that has been classified as a natural park since 1987. The lagoon hosts a diverse and abundant fish community and other species of commercial importance. Several economical activities are supported by shipping, and as such, vessel traffic within the Ria Formosa lagoon is very intense at some locations during particular seasons of the year, creating high levels of underwater noise. Recently, strong efforts are being made to turn the main inlet of the lagoon, the Faro-Olhão Inlet, a testing site for small scale tidal stream turbines, which will bring an additional source of underwater noise. Underwater noise can be one of a number of factors causing habitat degradation, as it can perturb fish behavior and cause physiological damage. Therefore, in order to comply with underwater noise pollution regulations, tidal energy technology developers are very interested in minimising the introduction of acoustic energy in the environment during the operation of their devices. Under the scope of project SCORE, which involved the deployment and operation of a floating tidal energy converter, this paper presents and discusses the first baseline noise monitoring performed at Ria Formosa. The acoustic data were collected in two occasions over several days, one in the winter and the other in the summer, in 2017. The obtained analysis results highlight the potential impact of the intense boat traffic in Ria Formosa, and the wide range of sound levels introduced in that ecosystem, and the high diurnal and seasonal variability.
