dc.description.abstractTransthyretin (TTR) is one of three thyroxine binding proteins found in the serum of higher animais. It has been shown to be synthesised and secreted in the choroid plexus of placental mammals, birds and reptiles but not in amphibians. Recently has cloned transthyretin from a sea bream liver cDNA library (Santos & Power, 1998). The objective of the present study was to determine the distribution of TTR in sea bream and study its regulation thyroid hormone. Ti and T4 (0,1 mg/g body weight) or saline was administered to sea bream by a single intraperitoneal injection. At 6h, I2h and 24h after injection treated and controls fish were killed and tissue removed. Expression of TTR was determined by RT-PCR, using homologous primers coupled with Southern blotting. The identity of PCR products was also confirmed by partial sequencing. TTR was expressed in liver, brain, heart, gill and gut and undetectable in kidney and muscle. Treatment with T3 e T4 changed the levei of expression of TTR in tissue. During the timescale studied there was a different up-regulation ofTTR expression by T4and T3 in liver and brain.por
dc.description.abstractA transretina (TTR) Ć© uma das trĆŖs proteĆ­nas que transporta a tiroxina no plasma sanguĆ­neo de animais superiores. VĆ”rios estudos demonstraram a sua sĆ­ntese e secreĆ§Ć£o no choroid plexus de mamĆ­feros placentĆ rios, aves, rĆ©pteis. Nos anfĆ­bios tal nĆ£o se verifica. Recentemente foi clonado o gene para a transretina a partir de um banco de cDNA de fĆ­gado de dourada (Santos & Power, 1998). O objectivo deste estudo foi determinar a distribuiĆ§Ć£o da transretina em alguns tecidos da dourada e estudar a influĆŖncia das hormonas produzidas pela tirĆ³ide na regulaĆ§Ć£o da transretina. Juvenis de dourada foram injectados intraperitonalmente com e T4 (0,1 mg/gr. peso corporal) ou com soluĆ§Ć£o salina. Seis horas, doze horas e vinte e quatro horas apĆ³s a injecĆ§Ć£o, peixes tratados (com Ti e T4) e controlos foram mortos e os tecidos removidos. A expressĆ£o da transtretina foi determinada por RT-PCR, usando primers homĆ³logos, e por Southern blot. A identidade dos produtos de PCR foi confirmada por sequenciaĆ§Ć£o. VerifĆ­cou-se, neste estudo que a transretina Ć© expressa no fĆ­gado, cĆ©rebro, coraĆ§Ć£o, brĆ¢nquias e intestino e indectĆ”vel no rim e mĆŗsculo. O tratamento com T3 e T4 muda os nĆ­veis de expressĆ£o da transretina nos tecidos. Durante os intervalos de tempo estudados verifĆ­cou-se que a expressĆ£o da transretina Ć© regulada de forma diferente no fĆ­gado e no cĆ©rebro, por acĆ§Ć£o da T3 e T4.por
