- Genetic differentiation of Diplodus sargus (Pisces: Sparidae) populations in the south-west MediterraneanPublication . A.Allozyme analysis of tissue samples of 1249 white sea bream Diplodus sargus from five localities of the south-west Mediterranean revealed a high degree of genetic polymorphism. The observed heterozygosity ranged from 0.4182 (Cape of Palos) to 0.3138 (Tabarca). Several populations were characterized by unique alleles. Examination of the spatial structure was performed using Nei’s distances and F-statistics, and indicated genetic differences between groups. One group, which clustered Tabarca and Guardamar, could be explained by the small geographical distance between them. Mazarrón and Cape of Palos samples showed genetic divergence from other samples (Guardamar, Tabarca and Águilas) and this difference may be as a result of local current systems and larval dispersal.
- Genetic and oceanographic tools reveal high population connectivity and diversity in the endangered pen shell Pinna nobilisPublication . We evaluate connectivity of a Mediterranean sessile species, Pinna nobilis. To address a large geographical scale, partial sequences of cytochrome oxidase I (COI, 590 bp) were used to evaluate phylogeographical patterns in the Western Mediterranean, and in the whole basin using overlapping sequences from the literature (243 bp). Additionally, we combined (1) larval trajectories based on oceanographic currents and early life-history traits and (2) 10 highly polymorphic microsatellite loci collected in the Western Mediterranean. COI results provided evidence for high diversity and low inter-population differentiation. Microsatellite genotypes showed increasing genetic differentiation with oceanographic transport time (isolation by oceanographic distance (IBD) set by marine currents). Genetic differentiation was detected between Banyuls and Murcia and between Murcia and Mallorca. However, no genetic break was detected between the Balearic populations and the mainland. Migration rates together with numerical Lagrangian simulations showed that (i) the Ebro Delta is a larval source for the Balearic populations (ii) Alicante is a sink population, accumulating allelic diversity from nearby populations. The inferred connectivity can be applied in the development of MPA networks in the Western Mediterranean.
- Phylogeography of the Atlanto-Mediterranean sea cucumber Holothuria (Holothuria) mammata: the combined effects of historical processes and current oceanographical patternPublication . H.; Gonzalez-Wanguemert, Mercedes; Marcos, C.; Perez-Ruzafa, A.We assessed the genetic structure of populations of the widely distributed sea cucumber Holothuria (Holothuria) mammata Grube, 1840, and investigated the effects of marine barriers to gene flow and historical processes. Several potential genetic breaks were considered, which would separate the Atlantic and Mediterranean basins, the isolated Macaronesian Islands from the other locations analysed, and the Western Mediterranean and Aegean Sea (Eastern Mediterranean). We analysed mitochondrial 16S and COI gene sequences from 177 individuals from four Atlantic locations and four Mediterranean locations. Haplotype diversity was high (H = 0.9307 for 16S and 0.9203 for COI), and the haplotypes were closely related (p = 0.0058 for 16S and 0.0071 for COI). The lowest genetic diversities were found in the Aegean Sea population. Our results showed that the COI gene was more variable and more useful for the detection of population structure than the 16S gene. The distribution of mtDNA haplotypes, the pairwise FST values and the results of exact tests and AMOVA revealed: (i) a significant genetic break between the population in the Aegean Sea and those in the other locations, as supported by both mitochondrial genes, and (ii) weak differentiation of the Canary and Azores Islands from the other populations; however, the populations from the Macaronesian Islands, Algarve and West Mediterranean could be considered to be a panmictic metapopulation. Isolation by distance was not identified in H. (H.) mammata. Historical events behind the observed findings, together with the current oceanographic patterns, were proposed and discussed as the main factors that determine the population structure and genetic signature of H. (H.) mammata
- The taxonomic status of some Atlanto-Mediterranean species in the subgenus Holothuria (Echinodermata: Holothuroidea: Holothuriidae) based on molecular evidencePublication . Borrero-Pérez, Giomar Helena; Pérez-Ruzafa, Angel; Marcos, Concepción; González-Wangüemert, MercedesMolecular and morphological data were used to evaluate the taxonomic status of the species Holothuria tubulosa Gmelin, 1790, Holothuria stellati Delle Chiaje, 1823, Holothuria mammata Grube, 1840, and Holothuria dakarensis Panning, 1939, belonging to the nominate subgenus Holothuria (Holothuria) (family Holothuriidae) from the Mediterranean Sea and Atlantic Ocean. A 16S rRNA marker distinguished three well-supported clades with clear genetic differentiation amongst them. The morphometric characters, although they reflected the clades, showed great variability, and some specimens from different clades overlapped. The morphological data and the literature suggest that the clades correspond to H. dakarensis (from Cape Verde Islands), H. mammata (from the Atlanto- Mediterranean area) and H. tubulosa (from the Mediterranean Sea). Holothuria stellati is considered here to be a junior subjective synonym of H. tubulosa. Great morphological intraspecific variation within H. tubulosa and H. mammata explains the confusion in the literature. Holothuria tubulosa includes specimens with distinctive ossicles, but others are similar to H. mammata. In these cases, the presence or absence of Cuvierian tubules proved a reliable indicator to the identity of these species; unfortunately this character is difficult to assess in preserved material. According to the results of discriminant analysis we propose a set of ossicle morphometric variables that permit the optimum assignation of individuals to the clades. Our results present a new perspective on the taxonomic status of species in Holothuria (Holothuria), and show how a molecular approach, combined with a morphological approach, can solve taxonomic problems.
- High gene flow promotes the genetic homogeneity of the fish goby Pomatoschistus marmoratus (Risso, 1810) from Mar Menor coastal lagoon and adjacent marine waters (Spain)Publication . Vergara-Chen, Carlos; González-Wangüemert, Mercedes; Marcos, Concepción; Pérez-Ruzafa, ÁngelThe extreme environmental variability of coastal lagoons suggests that physical and ecological factors could contribute to the genetic divergence among populations occurring in lagoon and open-coast environments. In this study we analysed the genetic variability of lagoon and marine samples of the sand goby, Pomatoschistus marmoratus (Risso, 1810) (Pisces: Gobiidae), on the SW Spain coast. A fragment of mitochondrial DNA control region (570 bp) was sequenced for 196 individuals collected in five localities: Lo Pagan, Los Urrutias and Playa Honda (Mar Menor coastal lagoon), and Veneziola and Mazarro´n (Mediterranean Sea). The total haplotype diversity was h = 0.9424 ± 0.0229, and the total nucleotide diversity was p = 0.0108 ± 0.0058. Among-sample genetic differentiation was not significant and small-scale patterns in the distribution of haplotypes were not apparent. Gene flow and dispersal-related life history traits may account for low genetic structure at a small spatial scale. The high genetic diversity found in P. marmoratus increases its potential to adapt to changing conditions of the Mar Menor coastal lagoon.
- Sea cucumbers, Holothuria arguinensis and H-mammata, from the southern Iberian Peninsula: variation in reproductive activity between populations from different habitatsPublication . Here, the reproductive biology of populations the two species was investigated in the southern Iberian Peninsula. Different populations located along a narrow latitudinal range displayed the same general reproductive pattern of summer-autumn spawning. However, significant differences in size, gonadal production and maturity profile between locations suggests the influence of site-specific factors. In Sagres and Ria Formosa H. arguinensis individuals were larger and had larger gonads than in Olhos de Agua, which had relatively more immature animals. The spawning and active gametogenesis periods were also longer in Sagres, possibly linked to specificity of food availability and tidal conditions. Ria Formosa also had larger H. mammata individuals with larger gonads than in Murcia and Olhos de Agua, possibly reflecting differences in feeding activity in different substrates (muddy/sandy vs rocky). Gametogenesis in H. arguinensis may be triggered by decreasing photoperiod and temperature, and spawning by increasing temperature. Altogether, these results, which include fecundity and size at first maturity, provide an important basis for the scientific management of sea cucumber fisheries in the region. (C) 2017 Elsevier B.V. - Molecular systematics of the genus Holothuria in the Mediterranean and Northeastern Atlantic and a molecular clock for the diversification of the Holothuriidae (Echinodermata: Holothuroidea)Publication . Borrero-Pérez, Giomar Helena; Gómez-Zurita, Jesús; González-Wangüemert, Mercedes; Marcos, Concepción; Pérez-Ruzafa, AngelThis work investigates the systematics of the genus Holothuria in the Mediterranean Sea and Northeastern Atlantic in the light of a wider molecular phylogenetic hypothesis of Holothuriidae, and it also provides a time-scale for the family diversification using mitochondrial markers and the molecular clock hypothesis. The subgenera Holothuria and Roweothuria are retrieved as paraphyletic. At least four separate lineages, with quite different time frameworks were identified. There are at least three species with an apparent long evolutionary history, H. forskali, H. sanctori and H. impatiens and six species belonging to Holothuria, Roweothuria and Vaneyothuria, which have diverged relatively recently.
