- Drug resistance associated genetic polymorphisms in Plasmodium falciparum and Plasmodium vivax collected in Honduras, Central AmericaPublication . The aim of this study was to determine the proportion of resistance associated genetic polymorphisms in P. falciparum and P. vivax collected in Honduras. Methods: Blood samples were collected from patients seeking medical attention at the Hospital Escuela in Tegucigalpa from 2004 to 2006 as well as three regional hospitals, two health centres and one regional laboratory during 2009. Single nucleotide polymorphisms in P. falciparum chloroquine resistance transporter (pfcrt), multidrug resistance 1 (pfmdr1), dihydrofolate reductase (pfdhfr) and dihydropteroate synthase (pfdhps) genes and in P. vivax multidrug resistance 1 (pvmdr1) and dihydrofolate reductase (pvdhfr) genes were detected using PCR based methods. Results: Thirty seven P. falciparum and 64 P. vivax samples were collected. All P. falciparum infections acquired in Honduras carried pfcrt, pfmdr1, pfdhps and pfdhfr alleles associated with chloroquine, amodiaquine and sulphadoxine-pyrimethamine sensitivity only. One patient with parasites acquired on a Pacific Island had pfcrt 76 T and pfmdr1 86Y alleles. That patient and a patient infected in West Africa had pfdhfr 51I, 59 R and 108 N alleles. Pvmdr1 976 F was found in 7/37 and two copies of pvmdr1 were found in 1/37 samples. Pvdhfr 57 L + 58 R was observed in 2/57 samples. Conclusion: The results indicate that P. falciparum from Honduras remain sensitive to chloroquine and sulphadoxine-pyrimethamine. This suggests that chloroquine and sulphadoxine-pyrimethamine should be efficacious for treatment of uncomplicated P. falciparum malaria, supporting current national treatment guidelines. However, genetic polymorphisms associated with chloroquine and sulphadoxine-pyrimethamine tolerance were detected in local P. vivax and imported P. falciparum infections. Continuous monitoring of the prevalence of drug resistant/tolerant P. falciparum and P. vivax is therefore essential also in Honduras.

- Influence of environmental and fishery parameters on loggerhead sea turtle by-catch in the longline fishery in the Azores archipelago and implications for conservationPublication . Ferreira, R. A.; Erzini, KarimOceanic juvenile loggerhead sea turtles, Caretta caretta, foraging in Azorean waters, are incidentally caught in drifting longlines that target swordfish, Xiphias gladius, and blue shark, Prionace glauca. Data were recorded during a longline gear modification experiment conducted in 2000. The results show that the captures are not uniformly distributed in relation to fishing effort, with 81% of the loggerheads (N = 232) caught in one-third of the sets (N = 93). The highest turtle catch rates were around the eastern group of islands, an area characterized by the presence of mesoscale eddies induced by bathymetric features. High catch rates coincided with the presence of the thermohaline front, between August and October, and when targeting blue shark. To investigate the causes of these differences we fitted a general linear model to evaluate the influence of mean depth, minimum depth, sea surface temperature (SST), soak area, soak time, moon phase and wind force on turtle and target species catch rates. These results revealed that mean depth and SST were most important for loggerhead catch rates, while minimum depth was the only significant variable for swordfish and mean depth, SST, wind force, moon phase and soak time influenced blue shark catches. The distribution of loggerhead sea turtles can be predicted by monitoring fishing areas in relation to bathymetry and SST. The impact of longline fishing could be greatly and quickly reduced by regulating the blue shark fishery and by increasing fishermen awareness for sea turtle conservation.

- Optical frequency tripling with improved suppression and sideband selectionPublication . Thakur, M. P.; Medeiros, Maria Carmo; Laurêncio, Paula; Mitchell, J. E.A novel optical dispersion tolerant millimetre-wave radio-over-fibre system using optical frequency tripling technique with enhanced and selectable sideband suppression is demonstrated. The implementation utilises cascaded optical modulators to achieve either an optical single sideband (OSSB) or double sideband-suppressed carrier (DSB-SC) signal with high sideband suppression. Our analysis and simulation results indicate that the achievable suppression ratio of this configuration is only limited by other system factors such as optical noise and drifting of the operational conditions. The OSSB transmission system performance is assessed experimentally by the transport of 4 WiMax channels modulating a 10 GHz optical upconverted RF carrier as well as for optical frequency doubling and tripling. The 10 GHz and tripled carrier at 30 GHz are dispersion tolerant resulting both in an average relative constellation error (RCE) of -28.7 dB after 40 km of fibre. (C)2011 Optical Society of America
- Plasmodium falciparum population dynamics during the early phase of anti-malarial drug treatment in Tanzanian children with acute uncomplicated malariaPublication . P.; Bjorkman, Anders; Martensson, AndreasBackground: This study aimed to explore Plasmodium falciparum population dynamics during the early phase of anti-malarial drug treatment with artemisinin-based combination therapy in children with clinical malaria in a high transmission area in Africa. Methods: A total of 50 children aged 1-10 years with acute uncomplicated P. falciparum malaria in Bagamoyo District, Tanzania, were enrolled. Participants were hospitalized and received supervised standard treatment with artemether-lumefantrine according to body weight in six doses over 3 days. Blood samples were collected 11 times, i.e. at time of diagnosis (-2 h) and 0, 2, 4, 8, 16, 24, 36, 48, 60 and 72 h after initiation of treatment. Parasite population dynamics were assessed using nested polymerase chain reaction (PCR)-genotyping of merozoite surface protein (msp) 1 and 2. Results: PCR-analyses from nine sequential blood samples collected after initiation of treatment identified 20 and 21 additional genotypes in 15/50 (30%) and 14/50 (28%) children with msp1 and msp2, respectively, non-detectable in the pre-treatment samples (-2 and 0 h combined). Some 15/20 (75%) and 14/21 (67%) of these genotypes were identified within 24 h, whereas 17/20 (85%) and 19/21 (90%) within 48 h for msp1 and msp2, respectively. The genotype profile was diverse, and varied considerably over time both within and between patients, molecular markers and their respective families. Conclusion: PCR analyses from multiple blood samples collected during the early treatment phase revealed a complex picture of parasite sub-populations. This underlines the importance of interpreting PCR-outcomes with caution and suggests that the present use of PCR-adjustment from paired blood samples in anti-malarial drug trials may overestimate assessment of drug efficacy in high transmission areas in Africa. - Comunicação no ensino-aprendizagem da matemática: práticas no 1.º ciclo do ensino básicoPublication . Guerreiro, António; Serrazina, Maria de Lurdes MarquêsO presente estudo analisa a evolução das conceções e práticas de comunicação matemática do professor do 1.º ciclo do ensino básico, no decorrer de um trabalho de natureza colaborativa focado na reflexão sobre as práticas de comunicação em sala de aula. O quadro teórico incide no conhecimento profissional do professor, com especial referência às suas formas paradigmáticas e narrativas, e na comunicação no processo de ensino-aprendizagem da matemática, com significativa incidência na comunicação matemática como interação social na sala de aula. O estudo enquadra-se numa metodologia qualitativa, adota o paradigma interpretativo, segue o design de estudo de caso e integra uma vertente de trabalho de natureza colaborativa entre o investigador e três professoras do 1.º ciclo do ensino básico. A recolha de dados incluiu as técnicas de observação, inquirição e colaboração. A análise de dados, organizada em estudos de caso, interpreta a plenitude da informação recolhida, através de referências significativas, reconstruindo a complexidade das vivências dos participantes nesta investigação. Os resultados do estudo apontam para a mudança das conceções sobre comunicação matemática através do reconhecimento da partilha comunicativa como fator determinante da aprendizagem da matemática. O reforço das interações entre os alunos suscita uma comunicação reflexiva e instrutiva, motivando a centralidade das aprendizagens nos processos comunicativos e no conhecimento partilhado entre os alunos e o professor. A assunção da singularidade dos conhecimentos dos alunos determina a alteração do foco das interações entre o professor e os alunos, reforçando a real inquirição dos alunos e a negociação dos significados matemáticos. A reflexão sobre as práticas de comunicação, apoiadas pelo visionamento das aulas e o sentido crítico do investigador, num clima de confiança e à-vontade, desencadeou um questionamento sobre as práticas de sala de aula e sobre o conhecimento profissional dos professores.
- Age and growth of the bigeye thresher shark, Alopias superciliosus, from the pelagic longline fisheries in the tropical northeastern Atlantic Ocean, determined by vertebral band countsPublication . Fernandez-Carvalho, Joana; Coelho, Rui; Erzini, Karim; Santos, Miguel N.The bigeye thresher, Alopias supercilious, is commonly caught as bycatch in pelagic longline fisheries targeting swordfish. Little information is yet available on the biology of this species, however. As part of an ongoing study, observers sent aboard fishing vessels have been collecting set of information that includes samples of vertebrae, with the aim of investigating age and growth of A. supercilious. A total of 117 A. supercilious were sampled between September 2008 and October 2009 in the tropical northeastern Atlantic, with specimens ranging from 101 to 242 cm fork length (FL) (176 to 407 cm total length). The A. supercilious vertebrae were generally difficult to read, mainly because they were poorly calcified, which is typical of Lamniformes sharks. Preliminary trials were carried out to determine the most efficient band enhancement technique for this species, in which crystal violet section staining was found to be the best methodology. Estimated ages in this sample ranged from 2 to 22 years for females and 1 to 17 years for males. A version of the von Bertalanffy growth model (VBGF) re-parameterised to estimate L0, and a modified VBGF using a fixed L0 were fitted to the data. The Akaike information criterion (AIC) was used to compare these models. The VBGF produced the best results, with the following parameters: Linf = 293cm FL, k =0.06/yr and L0 = 111 cm FL for females; Linf = 206 cm FL, k = 0.18/yr and L0 = 93 cm FL for males. The estimated growth coefficients confirm that A. supercilious is a slow-growing species, highlighting its vulnerability to fishing pressure. It is therefore urgent to carry out more biological research to inform fishery managers more adequately and address conservation issues.
- Driving south: a multi-gene phylogeny of the brown algal family Fucaceae reveals relationships and recent drivers of a marine radiationPublication . A.Understanding the processes driving speciation in marine ecosystems remained a challenge until recently, due to the unclear nature of dispersal boundaries. However, recent evidence for marine adaptive radiations and ecological speciation, as well as previously undetected patterns of cryptic speciation is overturning this view. Here, we use multi-gene phylogenetics to infer the family-level evolutionary history of Fucaceae (intertidal brown algae of the northern Pacific and Atlantic) in order to investigate recent and unique patterns of radiative speciation in the genus Fucus in the Atlantic, in contrast with the mainly monospecific extant genera. Results We developed a set of markers from 13 protein coding genes based on polymorphic cDNA from EST libraries, which provided novel resolution allowing estimation of ancestral character states and a detailed reconstruction of the recent radiative history. Phylogenetic reconstructions yielded similar topologies and revealed four independent trans-Arctic colonization events by Fucaceae lineages, two of which also involved transitions from hermaphroditism to dioecy associated with Atlantic invasions. More recently, reversion of dioecious ancestral lineages towards hermaphroditism has occurred in the genus Fucus, particularly coinciding with colonization of more extreme habitats. Novel lineages in the genus Fucus were also revealed in association with southern habitats. These most recent speciation events occurred during the Pleistocene glaciations and coincided with a shift towards selfing mating systems, generally southward shifts in distribution, and invasion of novel habitats. Conclusions Diversification of the family occurred in the Late-Mid Miocene, with at least four independent trans-Artic lineage crossings coincident with two reproductive mode transitions. The genus Fucus arose in the Pliocene but radiated within a relatively short time frame about 2.5 million years ago. Current species distributions of Fucus suggest that climatic factors promoted differentiation between the two major clades, while the recent and rapid species radiation in the temperate clade during Pleistocene glacial cycles coincided with several potential speciation drivers.
- Microplate-based high throughput screening procedure for the isolation of lipid-rich marine microalgaePublication . Pereira, Hugo; Barreira, Luísa; Mozes, Andre; Florindo, C.; Polo, Cristina; Duarte, Catarina V.; Custódio, Luísa; Varela, J.Abstract We describe a new selection method based on BODIPY (4,4-difluoro-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-s-indacene) staining, fluorescence activated cell sorting (FACS) and microplate-based isolation of lipid-rich microalgae from an environmental sample. Our results show that direct sorting onto solid medium upon FACS can save about 3 weeks during the scale-up process as compared with the growth of the same cultures in liquid medium. This approach enabled us to isolate a biodiverse collection of several axenic and unialgal cultures of different phyla.
