  • Setting preliminary biometric baselines for new target sea cucumbers species of the NE Atlantic and Mediterranean fisheries

    Commercial interest on sea cucumber species from the NE Atlantic and the Mediterranean is increasing.Holothuria polii, Holothuria tubulosa, Holothuria mammata and Holothuria arguinensis are the new targetspecies. The scarce biological and ecological knowledge on these species along their geographical distri-bution, is a major problem for their fisheries management. To address it, we carried out a morphometricstudy to describe the baseline population size structure, focused on the length–weight relationship andsize–weight distribution.In the NE Atlantic, the largest mean size and heaviest mean weight of H. mammata and H. arguinensiswere found in localities with upwelling, but another factors could be influencing on these results. In theMediterranean, Girona and Mallorca showed the heaviest specimens of H. polii, H. tubulosa and H. mam-mata, and the smallest were registered in Crete and Kusadasi. Significant differences in length and weightbetween populations were found for each species. These results could be linked with local environmentalconditions. Size frequency distribution was multimodal for all species except for H. mammata; the weightfrequency distribution was only unimodal for H. polii and H. mammata.This study provides novel data, helpful for stock and population assessment which could support theimplementation of effective management for the European sea cucumber target species.
  • Do hatchery-reared sea urchins pose a threat to genetic diversity in wild populations?

    In salmonids, the release of hatchery-reared fish has been shown to cause irreversible genetic impacts on wild populations. However, although responsible practices for producing and releasing genetically diverse, hatchery-reared juveniles have been published widely, they are rarely implemented. Here, we investigated genetic differences between wild and early-generation hatchery-reared populations of the purple sea urchin Paracentrotus lividus (a commercially important species in Europe) to assess whether hatcheries were able to maintain natural levels of genetic diversity. To test the hypothesis that hatchery rearing would cause bottleneck effects (that is, a substantial reduction in genetic diversity and differentiation from wild populations), we compared the levels and patterns of genetic variation between two hatcheries and four nearby wild populations, using samples from both Spain and Ireland. We found that hatchery-reared populations were less diverse and had diverged significantly from the wild populations, with a very small effective population size and a high degree of relatedness between individuals. These results raise a number of concerns about the genetic impacts of their release into wild populations, particularly when such a degree of differentiation can occur in a single generation of hatchery rearing. Consequently, we suggest that caution should be taken when using hatchery-reared individuals to augment fisheries, even for marine species with high dispersal capacity, and we provide some recommendations to improve hatchery rearing and release practices. Our results further highlight the need to consider the genetic risks of releasing hatchery-reared juveniles into the wild during the establishment of restocking, stock enhancement and sea ranching programs.

    A single male individual of Callinectes sapidus (Crustacea, Decapoda, Portunidae) was collected on 6 December 2014 in the Segura River mouth in southeastern Spain. Information on collection site, DNA barcoding, and its present distribution and dispersal strategies are provided.
  • Environmental variables, habitat discontinuity and life history shaping the genetic structure of Pomatoschistus marmoratus

    Coastal lagoons are semi-isolated ecosystems exposed to wide fluctuations of environmental conditions and showing habitat fragmentation. These features may play an important role in separating species into different populations, even at small spatial scales. In this study, we evaluate the concordance between mitochondrial (previous published data) and nuclear data analyzing the genetic variability of Pomatoschistus marmoratus in five localities, inside and outside the Mar Menor coastal lagoon (SE Spain) using eight microsatellites. High genetic diversity and similar levels of allele richness were observed across all loci and localities, although significant genic and genotypic differentiation was found between populations inside and outside the lagoon. In contrast to the FST values obtained from previous mitochondrial DNA analyses (control region), the microsatellite data exhibited significant differentiation among samples inside the Mar Menor and between lagoonal and marine samples. This pattern was corroborated using Cavalli-Sforza genetic distances. The habitat fragmentation inside the coastal lagoon and among lagoon and marine localities could be acting as a barrier to gene flow and contributing to the observed genetic structure. Our results from generalized additive models point a significant link between extreme lagoonal environmental conditions (mainly maximum salinity) and P. marmoratus genetic composition. Thereby, these environmental features could be also acting on genetic structure of coastal lagoon populations of P. marmoratus favoring their genetic divergence. The mating strategy of P. marmoratus could be also influencing our results obtained from mitochondrial and nuclear DNA. Therefore, a special consideration must be done in the selection of the DNA markers depending on the reproductive strategy of the species.
  • Improving the fitness of Holothuria arguinensis larvae through different microalgae diets

    Development and design of adequate feeding protocols on sea cucumber larvae are necessary for improvements on larvae growth, survival and production of competent juveniles. This work assessed the Holothuria arguinensis larvae growth and survival, fed with three different single microalgae diets of Chaetoceros calcitrans, Isochrysis galbana and Tetraselmis chui, and two microalgae diets based on a combination of C. calcitrans and T. chuii (C:T 1:1) and C. calcitrans, T. chuii and I. galbana (C:T:I 1:1:1) during 2 months. Holothuria arguinensis larvae fed with T:C:I, showed higher survival and larger larval and stomach sizes than the ones fed with the other diets. Comparing the results obtained from the single diets, H. arguinensis larvae fed with I. galbana grew and survived better during the first 5 days. However, higher growth was obtained on the larvae fed with C. calcitrans from day 5 to day 12 since this day, larvae fed with T. chuii showed the largest size. These results could be related to the different microalgae size and morphology, in addition to the nutritional value of the different microalgae used. However, no competent larvae that metamorphosed into doliolaria were obtained during this experiment. The tanks used were transparent, which could be causing light stress to the larvae, since competent larvae were obtained in opaque production tanks in our aquaculture installations during the same time period. The combined diet of C. calcitrans, T. chuii and I. galbana (C:T:I) could improve the growth and larvae competency of H. arguinensis.
  • Genetic and oceanographic tools reveal high population connectivity and diversity in the endangered pen shell Pinna nobilis

    For marine meta-populations with source-sink dynamics knowledge about genetic connectivity is important to conserve biodiversity and design marine protected areas (MPAs). We evaluate connectivity of a Mediterranean sessile species, Pinna nobilis. To address a large geographical scale, partial sequences of cytochrome oxidase I (COI, 590 bp) were used to evaluate phylogeographical patterns in the Western Mediterranean, and in the whole basin using overlapping sequences from the literature (243 bp). Additionally, we combined (1) larval trajectories based on oceanographic currents and early life-history traits and (2) 10 highly polymorphic microsatellite loci collected in the Western Mediterranean. COI results provided evidence for high diversity and low inter-population differentiation. Microsatellite genotypes showed increasing genetic differentiation with oceanographic transport time (isolation by oceanographic distance (IBD) set by marine currents). Genetic differentiation was detected between Banyuls and Murcia and between Murcia and Mallorca. However, no genetic break was detected between the Balearic populations and the mainland. Migration rates together with numerical Lagrangian simulations showed that (i) the Ebro Delta is a larval source for the Balearic populations (ii) Alicante is a sink population, accumulating allelic diversity from nearby populations. The inferred connectivity can be applied in the development of MPA networks in the Western Mediterranean.
  • First report of the nutritional profile and antioxidant potential of Holothuria arguinensis, a new resource for aquaculture in Europe

    This work reports for the first time the nutritional profile and antioxidant potential of the edible sea cucumber Holothuria arguinensis from the North-eastern Atlantic. H. arguinensis has high levels of protein, with the amino acids profile dominated by alanine, glycine and proline and low lysine/arginine ratios. Its carbohydrate and energetic contents are also low as well as the total lipid levels, although its lipid profile is rich in polyunsaturated fatty acids (PUFA), especially arachidonic, eicosapentaenoic and docosahexaenoic acids. In addition, H. arguinensis has high levels of calcium. The water and ethanol extracts show ability to scavenge free radicals and to chelate copper and iron ions. Our results indicate that H. arguinensis has a balanced nutritional quality suitable for human consumption. In addition, it contains compounds with antioxidant potential; thus its intake can contribute for a healthy and well-balanced diet.
  • Spatial patterns of genetic diversity in Mediterranean eagle owl Bubo bubo populations

    Little information is available on the patterns of genetic connectivity in owls. We studied the genetic structure of the eagle owl Bubo bubo (Linnaeus, 1758) in southeastern Spain at two different spatial scales. Seven microsatellites previously described for this species were used, although only six loci amplified correctly. The observed low genetic variation could be explained by the short dispersal distance, high mortality rate and high degree of monogamy shown by this large nocturnal predator. As expected, the highest genetic isolation was detected in the geographically most isolated population. Significant genetic differentiation was found among study units separated by less than 50 km. The territorial analysis showed interesting connectivity patterns related with the gene flow and turnover rate of the breeding individuals. The lowest genetic diversity was found in the region with the largest population, which could imply incipient inbreeding.
  • Habitat associations and seasonal abundance patterns of the sea cucumber Holothuria arguinensis at Ria Formosa coastal lagoon (South Portugal)

    Sea cucumbers have an important economic value and high demand. Despite that, there is a lack of knowledge about their biology, ecology and habitat distribution patterns, which is very important for sea cucumber stock management, establishment and sizing of no-take zones, restocking actions and selection of grow-out areas. This work aimed to determine the density, abundance and habitat associations of Holothuria arguinensis for a better understanding of its distribution along the coastal lagoon Ria Formosa and to select suitable areas for grow-out. In the duration of a year, monthly visual censuses were performed in two locations at Ria Formosa along the intertidal zone. The number and length of H. arguinensis ' individuals found were registered, and the coverage of algae and seagrass was estimated. It was found that H. arguinensis was distributed along the lower intertidal zone, linked to Zostera noltii meadows on muddy and sandy bottoms. These areas showed the densest population and the largest sizes of H. arguinensis. However, during the warmer months, H. arguinensis seemed to migrate to deeper waters in the channels, to avoid exposition to high temperatures and solar/UV irradiance during low tides. Areas located in the lower intertidal zone with Z. noltii meadows on sand-muddy bottoms should be selected for H. arguinensis grow-out sites.
  • Epidemiology of skin ulceration disease in wild sea cucumber Holothuria arguinensis, a new aquaculture target species

    Interest in wildlife epidemiology has increased in recent years. The control of diseases is critical for the survival of natural populations of economically valuable species. The present study is the first investigation of the etiology and epidemiology of skin ulceration disease in the sea cucumber Holothuria arguinensis, a new target species for fisheries and aquaculture in Europe. Bacterial cultures and molecular techniques were used to characterize this disease in animals collected during a survey across Ria Formosa Natural Park coastal lagoon in southern Portugal. Vibrio gigantis and V. crassostreae, which were both originally identified as disease agents in cultured oysters, were the most commonly isolated species of bacteria. Given that both sampling areas from which symptomatic H. arguinensis were collected were close to open oyster aquaculture facilities, this raises the possibility of an opportunistic infection, perhaps secondary to a decreased immune response caused by biotic or abiotic factors. An increase in prevalence of skin ulceration disease during the warmer season suggests that solar radiation and desiccation due to air exposure during low tide could be a cause of abiotic stress in the lagoon. Distributions of abundance and sizes of H. arguinensis in affected areas showed highest morbidity rates in adults. High fishery pressures throughout the study period could also cause elevations in prevalence and incidence rate of this disease. Skin ulcerative disease is endemic in this coastal lagoon. Disease monitoring is thus essential for the development of a conservation program to ensure the sustainability of fisheries and protection of natural resources.
