- Acoustic detection of bubbles in a pond covered by the seagrass Cymodocea nodosaPublication . Felisberto, Paulo; Silva, J. P.; Silva, A. E.This paper describes two experiments conducted in a pond covered by the seagrass Cymodocea nodosa at the Aquaculture Research Station of the Portuguese Institute for the Sea and Atmosphere in Olhao, Portugal, aiming at developing acoustic methods to assess oxygen production of seagrasses. The first experiment was carried out in July covering two days, when warm water and high photosynthetic rates give a high probability of oxygen supersaturation in water. The second experiment was carried out in late October, covering a period of 10 days, when seagrass productivity was expected to be lower than in July given the low irradiance and photoperiod. In the July experiment the high attenuation of low frequency pulses and broadband water pump noise (< 20 kHz) in the afternoon is ascribed to bubbles formation during oxygen supersaturation conditions. This hypothesis is coherent with the significant increase of the backscattering level, as measured by an acoustic backscatter system operating at 0.5, 1, 2, 4 MHz. Both, the attenuation of low frequency signals and backscattering level are correlated with oxygen supersaturation in water as measured by an optode. In the October experiment, when only water pump noise was acquired, the acoustic variability that can be related to photosynthetic activity was much weaker, nevertheless the attenuation shows a diurnal pattern correlated with the dissolved oxygen. The results suggest a significant release of oxygen as bubbles during photosynthesis, and therefore the potential contribution of acoustic methods to assess oxygen production of seagrass ecosystems.

D.Spatio-temporal variability of the diet of sardine Sardina pilchardus off Portugal was examined through analysis of the stomach contents of fish collected every 14 d from the west and south of Portugal during 2003/2004. Dietary composition of the modal sardine length class was assessed by determining the frequency of occurrence and carbon content of identified prey, and these 2 parameters were combined to estimate a modified index of relative importance of prey (mIRI). The most important prey for sardines were zooplankton, comprising crustacean eggs, copepods, decapods, cirripedes and fish eggs, dinoflagellates and diatoms (particularly the toxin-producer genus Pseudo-nitzschia), which together accounted for >90% of the estimated dietary carbon. Dietary seasonality was similar for both areas, except that the contribution of phytoplankton was higher for fish from the west Portuguese coast, where upwelling events are stronger and recurrent during spring and summer months. The predominance of prey <750 μm in sardine diet suggests that filter feeding is the dominant feeding mode used in the wild. Feeding intensity was similar for both sexes and for fish of different length classes and was higher on the west coast than in the south, which is probably related to the higher productivity of the west coast. Although there was high inter-annual variability in feeding intensity, this parameter was highest for both areas during spring and winter months. Temporal variability in satellite-derived chlorophyll a matched the temporal variability in the dietary contribution by phytoplankton and of sardine feeding intensity, suggesting further investigation of the potential use of satellite-derived chlorophyll a data as a proxy for sardine feeding intensity.
- Effect of maternal fat reserves on the fatty acid composition of sardine (Sardina pilchardus) oocytesPublication . Garrido, Susana; Rosa, R.; Ben-Hamadou, Radhouan; Cunha, M. D.We compared the fatty acid (FA) composition of the muscle and gonads of female Iberian sardines with hydrated oocytes collected during the 2002/03 spawning season off southern Portugal (November and February) and off western Portugal (February). Sardine condition and total FA concentration in the muscle decreased between the two sampling dates, while the gonadosomatic index was similar between samples. Total monounsaturated FA concentrations in sardine gonads were different for the three samples while saturated and polyunsaturated FA concentrations were similar. Significant linear relations were found between FA concentrations in female muscle and oocytes, including eicosapentaenoic acid (EPA; 20:5n−3) and arachidonic acid (AA; 20:4n−6), both being essential for normal larval development. The concentration of docosahexaenoic acid (DHA; 22:6n−3) in oocytes was independent on muscle concentration, probably resulting from its selective transfer to the oocytes. The EPA/ DHA ratio was highly conserved in sardine tissues, while DHA/AA and EPA/AA ratios varied significantly between samples. These results indicate that the FA content of eggs produced by sardines varies throughout the spawning season, egg FA concentrations decreasing as females lose condition, and FA composition also shows spatial variability. Both types of variability may have a significant impact on egg quality, particularly on the amount of reserves available to larvae affecting their resistance to starvation, and the appropriate FA composition required for normal growth.

Quality programmes are applied in a day to day basis having direct implications for production management and final costs of product. Most common criteria used for larval quality assessment are morphometrics, condition factor, histometrical indices, lipid analysis, nucleic acid ratios, enzyme activity and stress tests.
- Larval and juvenile development of dusky grouper Epinephelus marginatus reared in mesocosmsPublication . Cunha, M. E.; Re, P.; Quental-Ferreira, H.; Gavaia, Paulo J.; Pousão-Ferreira, P.The larval development of the dusky grouper Epinephelus marginatus up to the benthic juvenile stage is described in detail to establish a reference for their larval identification. Development is described in terms of ontogenetic changes in morphology, growth, pigmentation, fin structure and skeletal structure. Larvae were reared in mesocosms at a mean temperature of 24·3◦ C, salinity of 36·5, dissolved oxygen of 6·4mg l−1 and pH of 8·2. Newly hatched larvae had an estimated total length (LT) of 2·3 mm. On the second day post hatching the yolk was almost fully absorbed with traces of the oil globule still present, the eyes were already pigmented and mouth and gut functional. At this stage the cranial skeletal elements for feeding and breathing (mouth and gills) and the pectoral-fin support were already present. About 50% of the observed larvae had food in their guts. Pigmentation was very characteristic, consisting of two large chromatophores visible on the edge of the primordial fin, close to the midpoint of the post-anal region of the body and over the midgut and hindgut and post-anal portion of the body. At 2·9mm LT the emergence of the second dorsal-fin spine, characteristic of the Epinephilinae, was clearly visible. The pre-flexion stage started in larva of 3·2mm LT. At 5·5mm LT the larvae possessed posterior preopercular angle spines, and the dorsal and pelvic spines presented serrated edges and were pigmented. The water surface-tension-related death of the yolk sac and pre-flexion larvae described in the rearing of several other grouper species did not occur during E. marginatus culture. Notochord flexion, with initial ossification of the caudal-fin supporting elements, started at 6·6mm LT. At this stage the major melanophores, preopercular, dorsal and pelvic spines and mandibular teeth were already present. Transformation of larvae into juveniles occurred when larvae averaged 13·8mm LT. Juveniles with a mean LT of 20·1mm started to settle and most of them were benthic with a mean LT of 26·8 mm.

E.; Quental, H.; Barradas, Ana; Pousão-Ferreira, P.; Cabrita, Elsa; Engrola, S.One of the major obstacles to propagating dusky grouper, Epinephelus marginatus, is the difficulty of rearing the early larvae. We have successfully raised dusky grouper larvae in mesocosms using a mixed diet of endogenous plankton grown in the rearing tank and an exogenous supply of Brachionus plicatilis and Artemia sp. Newly hatched larvae at an initial density of 1.3 ind.l-1 were stocked in partially shaded 3 m3 circular outdoor tanks during the summers of 2007 and 2008. Before introducing newly hatched larvae, the water was left for six days to promote plankton growth. Larval growth occurred at two different rates: i) a faster rate from first feeding to the beginning of metamorphosis, and ii) a slower rate at transformation. Survival at the beginning of metamorphosis was less than 10% (33 DPH) in 2007 and between 25 and 50% (25 DPH) in 2008. High mortalities were observed during larval transformation. The estimated minimum food requirement per grouper larvae increased more than 300% from the beginning of the notochord flexion to the beginning of metamorphosis. To meet such a high feeding requirement the number of larger prey organisms/copepods in the mesocosm should be eight times higher during this time period.

It has recently been adapted for aquaculture production since it is well accepted by consumers and reaches high commercial values. After the initial description of the ontogenic events of skeletogenesis, systematic evaluation of the incidence of skeletal malformations by several groups revealed high levels of skeletal deformities reaching up to 90% of the population.
- Spatio-temporal variability in fatty acid trophic biomarkers in stomach contents and muscle of Iberian sardine (Sardina pilchardus) and its relationship with spawningPublication . D.Temporal variation in the fatty acid (FA) composition of stomach contents of Iberian sardines was compared to the relative contribution to dietary carbon made by different prey types for Wsh from two areas of Portugal. The effect of the FA content of the diet on sardine muscle FA composition was also studied, aiming at (1)analysing if FA biomarkers can be used as a complementary technique for the study of sardine diet and (2) to relate spatial and temporal variations of prey FA content with sardine condition and reproduction. Significant spatial differences in the FA composition of sardine diet occurred with concentrations of n-3 polyunsaturated FA, namely eicosapentaenoic acid [EPA, 20:5n-3] and linolenic acid 18:3n-3, being signifcantly higher in the diet of sardines from the west coast, whilst the diet of sardines from the south coast was richer in monounsaturated fatty acids (MUFA), namely the carnivory biomarker oleic acid 18:1n-9. These results are in agreement with the higher contribution made by diatoms and dinoflagellates to the diet of sardines of the west coast. Spatial variation in sardine dietary FA was also detected in their muscle composition, specifically for EPA, and the eicosapentaenoic/docosahexaenoic acid and (n-3)/(n-6) ratios, which were higher in sardines from the west coast. No difference in FA composition was detected between sexes, and the seasonal variability in sardine total FA concentration was primarily related to the seasonality of spawning. Sardines accumulate high concentrations of FAs during the resting stage of reproduction when the feeding intensity is similar or lower to that observed during the spawning season. Additionally, sardines show a high selective retention of MUFA and polyunsaturated FA (PUFA) throughout the year except at the beginning of the spawning season, when these FAs are largely invested in the formation of the gonads. Therefore, temporal and regional diVerences of prey environments are strong enough to be reflected in fish body composition, namely on the accumulation of essential FAs, which can have a strong impact on reproduction success for this species.
- Understanding the individual role of fish, oyster, phytoplankton and macroalgae in the ecology of integrated production in earthen pondsPublication . Cunha, M. E.; Quental-Ferreira, H.; Parejo, A.; Gamito, Sofia; Ribeiro, L.; Moreira, Márcio; Monteiro, I; Soares, F.; Pousao-Ferreira, P.In order to demonstrate that IMTA can be profitable and a good alternative to regular semi-intensive fish mariculture production in earthen ponds three different production treatments with distinct combinations of trophic levels were designed: (i) a combination of fish, filter feeders, phytoplankton and macroalgae, (ii) fish, filter feeders and phytoplankton and (iii) fish, phytoplankton and macroalgae, to evaluate the role of each trophic level within an Integrated Multi-Trophic Aquaculture system (IMTA). Each treatment was carried out under semi-intensive conditions with two replicates, in a total of 6 earthen ponds of 500 m(2) surface and depth of 1.5 m. The results showed that fish, oyster, phytoplankton and macroalgae integrated aquaculture is a healthy sustainable production system for mariculture in earthen ponds, providing much more fish supply compared with the other two treatments. Ponds with filter feeders had significantly lower turbidity (Nephelometric Formazin unit (FNU) of 13 in the morning and 17 in the afternoon) when compared to ponds without filter feeders (16 FNU in the morning and 20 FNU in the afternoon) with increased light penetration throughout water column (61 and 55 cm transparency in ponds with filter feeders compared to 51 cm in ponds without filter feeders) and consequently higher photosynthetic activity with significantly higher dissolved oxygen (5.4 mg L in the morning and 6.7 mg L-1 in the afternoon in ponds with filter feeders compared to 5.3 mg L-1 in the morning and 6.4 mg L-1 in the afternoon in ponds without filter feeders) and carbon sequestration (0.50 and 0.53 mg L-1 8 h(-1) in ponds with filter feeders and 0.43 mg L-1 8 h(-1) in ponds without filter feeders). In the fish, filter feeder, phytoplanton and macroalgae IMTA treatment, phytoplankton played a crucial role because they increased DO levels, removed the excess of nutrients from animal excretion, and was used as food by the filter feeders. Almost as important is the presence of filter feeders since they control the density of the microalgae and particulate matter in the ponds contributing to a more constant level of DO and higher transparency of the water column. The increased transparency and pond fertilization by oyster excretion, resulted in higher proliferation of phytoplankton (chlorophyll a concentrations of 16.5 mu gL(-1) and 20.2 mu g L-1 in ponds with filter feeder and 13.3 mu g L-1 in ponds without filter feeder) with benefits not only for filter feeders themselves but also for the macroalgae. At the end there was higher water quality and higher savings (14% day(-1)) in the energy costs for pond aeration. Meagre, white seabream and flathead grey mullet enhance their performance in IMTA systems with the presence of filter feeders with food conversion rates (FCR) of 1.52 when compared with 2.46 in the regular semi-intensive system composed by fish, phytoplankton and macroalgae. Meagre grew significantly more in IMTA systems with controlled macroalgae while white seabream and flathead grey mullet enhance their performance in the presence of macroalgae. The results show that the fish, oyster, phytoplankton and macroalgae integrated production in earthen ponds is an improved system compared to the regular semi-intensive fish production. The enhanced water quality in these systems leads to improved fish performance and higher biomass production, and to reduction in the energy power used, contributing to greater profitability.
