- Desenvolvimento e análise de um plano de negócios para turismo náuticoPublication . Rodrigues, André Fernandes; Rebelo, Sandra Cristina FranciscoPerante a crise pandémica que se instaurou um pouco por todo o mundo, foram vários os mercados a adaptar-se às mais diversas formas de contactar com o cliente. Alguns segmentos de mercado, como por exemplo o turismo rural e até experiências como o aluguer de iates, beneficiaram com esta nova abordagem de um turismo mais restringido em termos de contacto social. Num mundo a reinventar-se, surgem novas necessidades dos consumidores, possibilitando a criação de novas oportunidades. É neste contexto que surge a ideia de negócio explorada neste projeto, um modelo de negócio que se compromete a dar resposta a uma falha no lado da oferta no mercado de iates em Portugal. Assim, nasce a necessidade de elaborar um plano de negócios, onde se estabelece e se desenvolve a ideia de negócio, os riscos associados e quais os resultados que se espera obter, de modo que o empreendedor obtenha uma visão do futuro. A elaboração deste projeto consistiu no desenvolvimento de um plano de negócio com vista à criação de uma empresa privada de prestação de serviços charter de iates. A elaboração do mesmo, apresenta-se como uma oportunidade de estudar e analisar o mercado, permitindo verificar qual o espaço que a empresa LuxCharter poderá ocupar no mercado. A viabilidade e sustentabilidade do projeto é estruturada com base em várias análises dos ambientes interno e externo e mediante projeções financeiras, evidenciam o seu potencial de criação de valor. Após o plano financeiro realizado, são conhecidas as projeções e indicadores que nos permitem efetuar uma avaliação do projeto em diversas vertentes. O projeto, com um período de vida de 5 anos e um investimento inicial de 21.240,25€, apresenta, num cenário mais provável, um valor atual líquido de 172.913,54€ e uma taxa interna de rendibilidade de 259%. Mesmo em cenários mais pessimistas, o projeto apresenta-se viável. Desta forma, a presente ideia de negócio assume-se como um projeto de baixo investimento com uma rendibilidade consideravelmente atrativa.
- Políticas sociais e de reinserção no sistema prisional Português: estudo de caso: Estabelecimento prisional de OlhãoPublication . Rosado, António Luís Mendes Gonçalves; Martins, João EduardoEste trabalho aborda o problema da reinserção de ex-reclusos do Estabelecimento Prisional de Olhão. O principal objetivo foi analisar as políticas sociais e de reinserção nesse estabelecimento prisional. A investigação empírica assentou num estudo de caso e foram realizadas 10 entrevistas semiestruturadas a reclusos de modo a analisar-se as suas caraterísticas sociodemográficas, o contexto familiar, a escola e o trajeto escolar, as experiências no mundo do trabalho, as vivências da reclusão e, por fim, a questão da reinserção social. Tentámos compreender os caminhos que levaram à prisão, as perspetivas dos reclusos face à sua estadia no estabelecimento prisional e os processos de reinserção social, captados estes também a partir das perspetivas dos reclusos. Os resultados permitem dizer quanto aos processos e dinâmicas de reinserção social, que os reclusos entrevistados manifestam nos seus discursos uma forte dificuldade na transição da vida da prisão para o mundo exterior. Do mundo “lá dentro” para o mundo “cá fora”, sendo que quanto maior o tempo passado na prisão em decorrência do tipo de pena, mais difícil se torna a reintegração na sociedade. São identificadas as dificuldades de inserção no mercado de trabalho, o rompimento dos laços familiares como algo que agrava as dificuldades de reinserção social, a estigmatização associada à condição de “ex-recluso” geradora de descriminação social e ainda um Estado Social deficitário no apoio social à reintegração social dos reclusos, quer seja pelas faltas no apoio ao acesso à habitação quando necessária, aos apoios à integração no mercado de trabalho, ao acesso ao apoio psicológico ou ainda no acesso aos apoios sociais, percebidos estes como insuficientes para levar uma existência condigna no mundo social fora da instituição total onde estiveram envolvidos e encarcerados.
- Prospeção de psilídeos (Hemiptera: Psylloidea) e seus parasitoides a Sul de PortugalPublication . Duarte, Beatriz Zarcos; Neto, Luís Miguel MascarenhasA luta biológica clássica é frequentemente usada para o controlo de surtos de pragas de grande impacto na agricultura e floresta, no entanto, deve ser suportada por uma intensa atividade de investigação que assegure o sucesso destes programas e evite efeitos secundários indesejados. Recentemente iniciou-se um plano de luta biológica para o controlo de Trioza erytreae (Hemiptera: Triozidae) com o himenóptero Tamarixia dryi. Porém o conhecimento sobre psilídeos (superfamília Psylloidea a que pertence T. erytreae) em Portugal é muito reduzido e, para a continuação das largadas com T. dryi neste programa, são necessários estudos sobre a praga e respetivo parasitoide em várias vertentes, entre elas, a diversidade de psilídeos e fauna auxiliar associada. Neste âmbito, foi iniciada uma prospeção no Algarve com o objetivo de inventariar os psilídeos presentes nesta região, nativos e introduzidos, determinar as suas plantas hospedeiras e averiguar a fauna auxiliar, em particular os seus parasitoides. Ao longo do trabalho, foram realizadas mais de 330 amostras, utilizando as técnicas das batidas e sweeping net bem como recolhas de material vegetal, em mais de 140 espécies de plantas. Comparando com os dados existentes na Psyl´list foram encontradas 24 plantas hospedeiras, das quais 3 detetadas pela primeira vez a nível nacional e 5 a nível mundial. No que diz respeito aos psilídeos encontrados durante a prospeção, pelo menos 32 morfoespécies foram identificadas, quase todas até ao género, e algumas até ao nível específico. Foram igualmente encontrados parasitoides em 7 destes psilídeos. Este é o primeiro estudo exaustivo realizado sobre psilídeos no Algarve o qual trouxe novos dados sobre a fauna desta superfamília, plantas hospedeiras e parasitoides. É um trabalho que requer, no entanto, uma continuação com o objetivo de proceder a uma identificação específica de todos os psilídeos encontrados e uma continuação e alargamento da prospeção iniciada.
- Potencial terapêutico da irisina na terapêutica da obesidadePublication . Nóbrega, Inês Sofia Pires; Serralheiro, Ana Isabel AzevedoA obesidade constitui uma doença crónica e um problema de saúde pública, que se caracteriza por um excesso de gordura corporal acumulada, que pode ser prejudicial à saúde de um indivíduo. Resulta de um conjunto de balanços energéticos positivos, em que o consumo calórico é superior ao gasto. A sua prevalência tem vindo a aumentar nos últimos anos, tendo quase triplicado nos últimos 40 anos, a nível mundial. A terapêutica farmacológica da obesidade é, atualmente, muito limitada, e inclui apenas três fármacos aprovados a nível europeu: o orlistato, o liraglutido e a associação de bupropiom e naltrexona. A terapêutica de 1ªlinha é o liraglutido, por ter menos efeitos adversos, maior evidência científica e maior efetividade. A terapêutica não farmacológica, que constitui a primeira opção de tratamento, inclui a terapêutica nutricional, a prática de exercício físico regular, a terapêutica comportamental, e em casos mais extremos, a cirurgia bariátrica. Em 2012, foi descoberta, em Harvard, uma hormona, a irisina, produzida pelo músculo esquelético durante a prática de exercício físico, capaz de aumentar a termogénese e de promover o browning do tecido adiposo, constituindo uma potencial opção na terapêutica da obesidade. Até ao momento, ainda não foi identificado um recetor específico para a irisina e o seu mecanismo de ação ainda não está completamente esclarecido. Esta mioquina surge, assim como uma forma de justificar os efeitos benéficos do exercício físico em múltiplas patologias. Para além da obesidade, existem estudos sobre a aplicação desta mioquina noutras doenças, quer como tratamento quer como biomarcador da sua evolução, nomeadamente na diabetes tipo 2, na doença cardiovascular, na doença de Alzheimer, entre outras. No entanto, os resultados obtidos pelos diversos estudos são, muitas vezes controversos. Além disso, a maioria dos estudos recorre a modelos animais, e nem sempre os resultados são transponíveis para a espécie humana.
- Evaluation of the antioxidant and anti-inflammatory activity in macroalgae asparagopsis armata and asparagopsis taxiformisPublication . Appiah, Amponsah Preko; Barreira, LuísaIn recent times, natural compounds have become widely employed in the treatment of human illnesses and health conditions such as cancer, neurological disorders, multiple sclerosis, diabetes, atherosclerosis, arthritis, and cardiovascular diseases all around the world. Plants have been the principal source of numerous pharmaceutical agents throughout history. However, in this modern era, much emphasis has been placed on the amazing biodiversity of life in the marine environment, which has proven to be an extraordinary repository of novel bioactive compounds with diverse structural and chemical properties, as well as a source of inspiration for new drug discovery. In the never-ending quest for novel safe and effective anti-inflammatory and antioxidant compounds beneficial to human health, this research focused on evaluating the ability of two distinct species of the macroalgae Asparagopsis spp (Asparagopsis taxiformis, as a gametophyte and as a sporophyte, and Asparagopsis armata as a gametophyte) to contribute for the treatment of anti-inflammatory diseases by assessing their antioxidant and anti-inflammatory activities. Ethanol, ethyl acetate and hexane were employed as solvents for the extraction of bioactive compounds from Asparagopsis spp. These solvents cover a wide range of polarity indices with polarity ranging from 0.1 for hexane, 4.4 for ethyl acetate, and 5.2 for ethanol to maximize the spectrum of extracted compounds. In this study, the best extraction yields were obtained with ethanol across all species of Asparagopsis with yields of 12.4% in A. armata, 9.0% in A. taxiformis as a gametophyte and 6.8% in the same as a sporophyte. Hexane yielded the least amount of extracts across all the species with yields of 0.4% in A. armata, 1.1% and 1.0% in A. taxiformis as a gametophyte and sporophyte respectively. Ethyl acetate showed yields of 2.4% in A. armata and 2.5% and 1.8% in A. taxiformis gametophyte and sporophyte respectively. Antioxidant properties were evaluated using tests such as ABTS, DPPH, iron & copper chelating activity, total phenolic content, and iron reducing activity while anti-inflammatory analysis was conducted specifically analysing for COX-2 inhibition. Results obtained suggests that Asparagopsis spp contain several bioactive compounds that exhibit good antioxidant abilities especially with metal chelating activities. Activity results in the DPPH assays were rather low (below 50% activity) across all species. For the ABTS assay, only the ethanolic extract of A. armata showed activity above 50% with an IC50 value of 0.24 mg/mL. The best results for copper chelating activity were observed in the hexane extracts of A. taxiformis sporophyte with an IC50 of 0.12 mg/mL. This was followed by the hexane extract of A. armata with an IC50 of 0.28 mg/mL. The best iron chelating activity was seen in the hexane extract of A. taxiformis sporophyte once again with an IC50 of 0.09 mg/mL. The ethyl acetate extracts of A. armata and A. taxiformis sporophyte also showed IC50 values of 0.25 and 0.24 mg/mL respectively. The iron reducing antioxidant assay showed good results for ethyl acetate extracts across all species with IC50 values of 0.07 mg/mL recorded of A. taxiformis sporophyte, 0.16 mg/mL for A. armata and 0.34 mg/mL for the gametophyte life stage of A. taxiformis. These extracts were observed to show activities higher than the gallic acid positive control. The macroalgae also demonstrated tremendous potential for COX-2 inhibition activity at an extract concentration of 1 mg/mL for all species (especially the hexane extracts showing over 50% activity across species) with the highest activity of 98.9 ± 0.4% observed in A. taxiformis gametophyte hexane extracts. Ethanol extract from A. taxiformis gametophyte also showed an activity of 79.1 ± 1.8% to take the second place whereas the hexane extract from A. taxiformis as a sporophyte showed inhibitory activity of 72.3 ± 5.1% taking the third spot. These activities compared to the ibuprofen positive control that was used at a concentration of 5 μg/mL and showed an activity of 55.0 ± 13.2% demonstrates that even as crude extracts Asparagopsis spp have great anti-inflammatory potentials and therefore in their highly refined states, extracts from Asparagopsis spp may be very competitive against other commercially known anti-inflammatory compounds on the market. Total phenolic content analysis (TPC) conducted on the extracts confirmed the presence of phenolic compounds in all the species of Asparagopsis evaluated. The highest levels of TPC were observed in the ethyl acetate extracts of all the species. Levels in the ethyl acetate extracts ranged from 1.50 ± 0.12 mg-GAE/g for A. armata, 1.19 ± 0.03 mg-GAE/g for A. taxiformis gametophyte to 0.61 ± 0.07 mg-GAE/g for A. taxiformis sporophyte. The relatively higher phenolic content observed in the ethyl acetate extracts suggests that most of the phenolic compounds present in Asparagopsis spp may be phenolic compounds of lower polarity such as tocopherols and tocotrienol which have been demonstrated to be well extracted by ethyl acetate. In general, the levels of total phenolic compounds present in Asparagopsis spp were determined to be relatively low compared to other macroalgae in literature. Despite the relatively low phenolic contents demonstrated in Asparagopsis spp it still showed good antioxidant and anti-inflammatory activities thus the antioxidant activity observed in the crude extracts were not solely dependent on the phenolic compounds but rather to a greater extent other bioactive compound(s) that may be present in the crude extracts of Asparagopsis spp. Statistical analysis in the form of ANOVA at a 95% Confidence Interval (alpha = 0.05) and Tukey multiple comparison test (P<0.05) were performed on the results obtained for all the antioxidant and anti-inflammatory tests conducted. The analysis revealed significant differences between the Asparagopsis species (A. taxiformis and A. armata) considered in this study and most importantly between the life stages of A. taxiformis as a gametophyte and a sporophyte. Multivariate data analyses in the form of principal component analysis (PCA) and cluster analysis (CA) were carried out between the antioxidant assay, metal chelating assays, COX-2 anti-inflammatory assay, and total phenolic content assay. Results of the analysis revealed that the solvent type used for extraction had a great influence on the antioxidant performance of the extracts. The analysis also explored the similarities between the tests and identified a good positive correlation between copper chelating activity test and COX-2 anti-inflammatory test. This observation is of great importance because COX-2 inhibition test is noted to be time consuming, complex, and very expensive to carry out. Hence in the event of a study where a large number of samples are to be tested it becomes challenging and puts a great strain on not only human resources, but financial resources and time as well. However, copper chelating activity (CCA) is comparatively simple and economical with time and funds hence could be a great way of pre-screening samples to determine which samples are likely to show good results with COX-2 analysis and hence reduce sample size which will eventually save time and other resources. Overall, findings presented from this research work suggests that the macroalgae Asparagopsis spp demonstrates high anti-inflammatory and antioxidant potentials for use in the pharmaceutical and nutraceutical industries.
- The effect of nest abundance of Loggerhead turtles on the nest predation rate of ghost crabs in Cabo VerdePublication . Boner, Kaja Lena; Vinagre, Catarina; Marco, AdolfoThe Loggerhead sea turtle (Caretta caretta) is facing intense nest predation globally. The subpopulation nesting on the Cabo Verde Archipelago, which is categorized as Endangered on the IUCN List, has been encountering high predation rates caused by ghost crabs (Ocypode cursor), the main predator on these oceanic islands. This study investigates the effect of nest abundance of Loggerheads on nest predation by ghost crabs on Boa Vista, Cabo Verde, from 2013 to 2021. Boa Vista hosts around 60% of the Loggerhead nests in Cabo Verde which is one of the world's largest Loggerhead rookeries. The nest abundance was recorded during night patrol and daily beach surveys at dawn. The predation of ghost crabs was evaluated using natural Loggerhead nests that were checked daily for any evidence of predation, inundation, erosion, or any other relevant disturbances to the nest. The clutch size of these nests was counted during oviposition and the nests were exhumed after the incubation period to assess the number of hatched, enclosed, and missing eggs. The interannual variability in Loggerhead nests was high, ranging from 1123 to 33496 nests. The nest mortality by crab predation was strongly negatively correlated to the nest abundance. Seasons with a low nest abundance showed high crab predation rates of up to 75%, whereas in seasons with high nest abundance crab predation rates as low as 4% were observed. Higher nest abundance might exceed the capacity to consume sea turtle eggs of the ghost crab population on the studied beach. Through predator satiation caused by high nest abundance, the risk of predation and the extent of the predation may be reduced. The findings give an interesting insight into this predator-prey interaction but also highlight the importance of conservation efforts in reducing predation, especially for areas and seasons with low sea turtle nest abundance.
- Synthesis, characterization and biological evaluation of new Zinc(II)-8-hydroxyquinoline derived complexesPublication . KWAO, SARAH; Correia, Isabel Maria Palma Antunes; Cavaco, Maria Isabel RodriguesMetallodrugs like cisplatin and related platinum compounds, widely used for the treatment of cancer patients, have severe side effects and acquired resistance, which are limiting their use. In addition, several cancer conditions are not responsive to available treatments and thus, viable clinical solutions are still lacking. Metal-based drugs have received a great deal of attention in recent years owing to their properties and benefits in biomedical, therapeutic and diagnostic systems. A wider understanding of the anticancer properties of transition metal complexes like Ru, V, Cu, and Zn and their interactions in biological media is very necessary to ensure their effectiveness and safety. In the current work, four Schiff base ligands derived from the condensation of 8-hydroxy-2-quinolinecarbaldehyde and benzohydrazides (H2L1-H2L4), and their corresponding zinc complexes, ZnL1, ZnL2, ZnL3, and ZnL4 were synthesized. All compounds were characterized by elemental analyses, FTIR, UV–Vis and NMR spectroscopies as well as ESI-MS spectrometry. The stability of the complexes was evaluated in DMSO and under physiological conditions (HEPES buffer (10 mM, pH 7.4) containing DMSO). The interaction between Schiff bases and their corresponding Zn(II) complexes with biological molecules, namely bovine serum albumin (BSA) was investigated using UV-Vis and fluorescence spectroscopies at 298 K. Fluorescence quenching of BSA upon interaction with the complexes was also analyzed at 298, 301, 305 and 310 K. Using the Stern-Volmer formalism, the binding constants indicated moderate to strong binding to albumin with values ranging from Kb = 0.63 to 7.81×104 M-1 for the first three complexes. These results reveal good binding propensity with the protein. Additionally, evaluation of thermodynamic parameters revealed that hydrophobic interactions are the main interactive forces involved in the zinc complexes binding to BSA. In vitro cytotoxic activity of the ligands and their complexes was evaluated on prostate carcinoma (LNCaP FGC), acute T-cell leukaemia (Jurkat E6-1), myeloma (MM.1S), and non-cancerous immortalized human HEK-293 cell lines. Strong cytotoxic activities of both ligands and complexes were observed against all three cancer cell lines. In the non-cancer cell line, the ligands exhibited lower IC50 values than their counterpart complexes. However, the metal complexes have solubility issues, making it difficult for their evaluation.Overall, the current ligand system shows high potential for cancer treatment due to the high selectivity towards the studied cancer cells.
- Turismo sustentável: análise do comportamento dos turistas portuguesesPublication . Almeida, Victor Augusto Couto Queiroz de; Santos, Margarida C.Esta dissertação aborda a temática do turismo sustentável, mais especificamente o comportamento dos viajantes e seu impacto para a sustentabilidade. Apesar de sua inegável importância para a economia e a sociedade, a atividade turística gera desequilíbrios ambientais, sociais e econômicos. O comportamento dos turistas está entre os principais promovedores dos referidos efeitos. O objetivo principal desta investigação consiste em conhecer o comportamento dos portugueses com relação às práticas sustentáveis, quando estes realizam viagens e no seu quotidiano. O estudo exploratório, caracterizado por um enquadramento teórico, busca aprofundar a temática da sustentabilidade e sua relação com o turismo, bem como examinar as práticas sustentáveis adotadas no quotidiano e no contexto das viagens. A pesquisa empírica, de caráter quantitativo, dá-se através da realização de um inquérito por questionário, contando com uma amostra não-probabilística de 172 indivíduos. Os dados recolhidos e analisados sugerem que os portugueses demonstram ampla familiaridade com o conceito de sustentabilidade e, de forma geral, adotam práticas sustentáveis no seu dia a dia. No contexto das viagens verifica-se um aumento dos níveis de consciência dos portugueses e uma mudança gradual em seu comportamento, mas com um grau de implementação de hábitos responsáveis inferior, se comparado ao quotidiano. Os resultados não revelam uma unanimidade, mas sim viajantes em diferentes estágios quanto à adoção de práticas sustentáveis, com uma tendência favorável. No entanto, as intenções nem sempre se traduzem em condutas conscientes, podendo haver uma lacuna entre as atitudes expressas e o comportamento real por parte dos viajantes portugueses, confirmando o que se verifica sobre o tema na literatura. Este estudo demonstra relevância para diversos stakeholders relacionados ao turismo em Portugal e contribui para o desenvolvimento de intervenções destinadas a aprimorar o comportamento sustentável dos turistas.
- The interconnectedness of decentralized finance with the traditional financial system and potential implications for systemic riskPublication . Gomes, Hugo Miguel de Araújo Veloso; Coelho, Luís; Neugebauer, KatjaDecentralized Finance (DeFi) was introduced to replicate the traditional financial system in a more open and transparent way by removing intermediaries and enabling peer-to-peer interaction in a way that was not possible until now. However, with innovation, unknown risks and concerns always arise, and the case of DeFi is no different. The main objectives of this research are as follows: to understand how the DeFi ecosystem works; examine if this new technology can bring innovation to the financial system; analyze if it can be a potential threat to the global financial system stability; and understand which aspects of interconnectedness can exist between DeFi and the traditional financial system, how prone can DeFi be to systemic risk, and how this could impact the global financial system. The analysis shows that DeFi has traits that have the potential to bring great innovations to the financial system, although it also has several risks. The study shows that some of the main characteristics of DeFi are already creating a bridge between both markets; the interconnectedness between both markets is growing with the growth of the financial institutions participating in the DeFi markets. Whenever the level of interconnectedness is high, the systems tend to be more prone to systemic risk. The case of DeFi is no different. Systemic risk is present within both markets, and potential spillovers are more likely to happen with the continuous growth of interconnectedness. Regulators and policy makers will need to explore and create new ways to enforce the regulations needed to create a space where users are protected and global financial stability is assured without compromising the innovation happening within this ecosystem.
